目的 检测肾癌(RCC)小鼠外周血(PB)、癌组织(TT)和癌旁组织(AT)中内皮祖细胞(EPCs),探讨其在RCC进展中的动员特征及机制.方法 雄性BALB/c裸鼠随机分为3组即RCC组、假手术组(Sham组)、正常组(Normal组).分别在移植肿瘤细胞后21、28、35、42、49 d处死小鼠,获取血和组织标本.评估循环内皮祖细胞(CEPCs)、血浆中血管内皮生长因子(VEGF)、基质细胞衍生因子-1(SDF-1)以及正常肾组织(NT)、假手术肾组织(ST)、AT和TT的EPCs、VEGF和SDF-1基因的表达.结果 与Normal以及Sham组比较,RCC组CEPCs和血浆VEGF、SDF-1表达水平明显升高(P<0.05),CEPCs和血VEGF、SDF-1之间呈正相关(r1=0.584,r2=0.579).AT中EPCs水平明显高于TT及NT(P <0.05),与CEPCs之间呈正相关(r=0.857).AT中VEGF和SDF-1表达显著高于TT及NT(P <0.05),且与血浆VEGF、SDF-1呈正相关(r1 =0.618,r2=0.421).结论 AT中VEGF和SDF-1在EPCs动员中具有重要作用.

목적 검측신암(RCC)소서외주혈(PB)、암조직(TT)화암방조직(AT)중내피조세포(EPCs),탐토기재RCC진전중적동원특정급궤제.방법 웅성BALB/c라서수궤분위3조즉RCC조、가수술조(Sham조)、정상조(Normal조).분별재이식종류세포후21、28、35、42、49 d처사소서,획취혈화조직표본.평고순배내피조세포(CEPCs)、혈장중혈관내피생장인자(VEGF)、기질세포연생인자-1(SDF-1)이급정상신조직(NT)、가수술신조직(ST)、AT화TT적EPCs、VEGF화SDF-1기인적표체.결과 여Normal이급Sham조비교,RCC조CEPCs화혈장VEGF、SDF-1표체수평명현승고(P<0.05),CEPCs화혈VEGF、SDF-1지간정정상관(r1=0.584,r2=0.579).AT중EPCs수평명현고우TT급NT(P <0.05),여CEPCs지간정정상관(r=0.857).AT중VEGF화SDF-1표체현저고우TT급NT(P <0.05),차여혈장VEGF、SDF-1정정상관(r1 =0.618,r2=0.421).결론 AT중VEGF화SDF-1재EPCs동원중구유중요작용.
Objective To investigate the endothelial progenitor cells (EPCs) in the peripheral blood (PB),tumor tissue (TT) and adjacent nonmalignant tissue(AT) of mice with renal cell carcinoma progression (RCC) and explore mobilized characteristic and mechanism in RCC.Methods Male,BALB/c nude mice were randomly divided into three groups:RCC group; sham group and normal group.Mice were killed respectively at days 21,28,35,42,49 after implant tumor cells,blood and tissue samples were harvested.Numbers of Circulating endothelial progenitor cells,plasma levels of vascular endothelial growth factor and stromal cell-derived factor-1,numbers of EPCs and mRNA (VEGF and SDF-1) expression in normal tissue (NT),sham tissue (ST),AT and TT were evaluated.Results CEPCs levels and plasma angiogenic factor (VEGF and SDF-1) were higher in PB of the RCC group than those in the normal and sham groups (P < 0.05)and there are a positive correlation between them (r1 =0.584,r2 =0.579).EPCs levels in AT were significantly higher than those in TT and NT (P < 0.05),which were positively correlated with CEPCs levels (r =0.857).VEGF and SDF-1 expression in AT was significantly higher compared to that in TT and NT (P < 0.05),which were positively correlated with that in plasma (r1 =0.618,r2 =0.421).Conclusion VEGF and SDF-1 in adjacent tissue play key role in entire mobilized process of EPCs.
