目的 探讨细胞外信号调节酶5(ERK5)与微小RNA-145(miR-145)在前列腺癌组织中表达及两者之间的关系.方法 分别采用免疫组织化学和原位杂交的方法检测组织芯片上40例前列腺癌患者80点前列腺癌组织以及15点周围正常组织中ERK5与miR-145的表达,并与临床病理特征进行统计学分析.结果 ERK5在前列腺癌组织中阳性表达率为77.5%,而在癌旁组织阳性表达率为26.7%,两者表达差异有统计学意义(P<0.05).miR-145在前列腺癌组织表达率为27.5%,在癌旁组织阳性表达率为62.5%,两者差异有统计学意义(P<0.05).ERK5表达与患者的Gleason评分、前列腺特异抗原(PSA)水平、临床分期明显相关(P<0.05).miR-145在Ⅱ期的前列腺癌患者的表达率明显高于Ⅲ期,ERK5与miR-145在前列腺癌中呈负相关.结论 ERK5与miR-145在前列腺癌中表达呈负相关,提示miR-145与ERK5在前列腺癌发生及进展中密切相关.

Objective To study the expression of extracellular signal-regulated kinase 5 (ERK5) and microRNA-145 (miR-145) in prostate cancer (PCa) and their correlation.Methods The expression of ERK5 and miR-145 in 80 cases of carcinoma tissues and 15 cases of normal tissues (40 cases of 95 cores in a set of tissue microarrays) was respectively measured by immunohistochemistry and in situ hybridization method.The potential relationship between ERK5 or miR-145 and clinicopathological features was also analyzed.Results ERK5 expression was significantly higher in carcinoma tissues (77.5%) than in normal tissues (26.7%).miR-145 expression was significantly lower in carcinoma tissues (27.5%) than in normal tissues.Overexpression of ERK5 in carcinoma tissues was positively correlated with prostate specific antigen,gleason scores and clinical stages.The lower expression of mirR145 in carcinoma tissues was inversely correlated with clinical stages between Ⅱ and Ⅲ.There was a strong inverse correlation between ERK5 and miR-145.Conclusion The expression of ERK5 and miR-145 in prostate cancer tissues has a strong inverse correlation,which prompts that ERK5 and miR-145 play an important role in prostate cancer development and progression.
