目的 探讨甲状腺乳头状癌(PTC)细胞株TPC-1、甲状腺鳞癌细胞株SW579、分化型甲状腺癌细胞株FTC-133以及PTC组织中卷曲螺旋结构域蛋白67(CCDC67)基因mRNA和蛋白表达的相关性.方法 采用逆转录-聚合酶链反应(RT-PCR)和Western blot技术,检测甲状腺癌细胞株以及56例PTC及对应癌旁组织中CCDC67基因mRNA和蛋白表达.结果 甲状腺癌细胞株中均无CCDC67基因mRNA和蛋白的表达.癌旁组织中均出现CCDC67 mRNA和蛋白的表达.PTC组织中CCDC67 mRNA的表达率为41.4% (23/56),蛋白的表达率为46.4%(26/56),两者均与患者的TNM分期、病理分级及淋巴结转移明显相关(P<0.05).CCDC67 mRNA与蛋白的表达明显相关(P<0.05).结论 CCDC67在甲状腺癌细胞株中表达缺失,CCDC67 mRNA及其蛋白在PTC组织中表达水平均低于癌旁组织,CCDC67基因在PTC的发生、发展中起着重要作用.

Objective To investigate the mRNA and protein expression of coiled-coil domain containing 67 (CCDC67) gene in papillary thyroid carcinoma (PTC).Methods Reverse transcription-polymerase reaction (RT-PCR) and Western blotting were used to detect the CCDC67 mRNA and protein levels in the cell lines of thyroid carcinoma and 56 human PTC specimens and their matched adjacent non-cancerous epithelium (NCE).Results There is no expression of CCDC67 in thyroid carcinoma cell lines.All cases had expression of CCDC67 mRNA and protein in adjacent normal tissues,but the expression of CCDC67 mRNA was down-regulated significantly,only 41.4% (23/56) in 56 cases of thyroid carcinoma appeared CCDC67 mRNA expression,it was correlated with the tumor TNM stage,pathological grade and lymph node metastasis (x2 =11.368,4.861,6.321,P < 0.05).The expression of CCDC67 protein was down-regulated significantly,only 46.4% (26/56) in 56 cases of thyroid carcinoma appeared CCDC67 protein expression,it was correlated with the tumor TNM stage,pathological grade and lymph node metastasis (x2 =4.229,4.314,5.540,P < 0.05).Furthermore there was a distinct correlation between the mRNA and protein expression of CCDC67 gene (x2 =25.775,P < 0.01).Conclusion There is no expression of CCDC67 in thyroid carcinoma cell lines.The expression of CCDC67 mRNA and protein was down-regulated in PTC.A lower expression of CCDC67 gene may play an important role in the carcinogenesis and progression of PTC.
