目的 研究高压氧(HBO)对人鼻咽癌细胞增殖与死亡的影响及其机制.方法 将实验培养的人鼻咽癌CNE2Z细胞分为对照组、HBO 1组(0.20 MPa)和HBO 2组(0.25 MPa).采用甲基噻唑基四唑(MTT)法分析细胞增殖抑制率,碘化丙啶(PI)染色观察细胞死亡情况,并采用黄嘌呤氧化酶法测定SOD的含量、硫代巴比妥法测定MDA的含量.结果 HBO 1组和HBO 2组的细胞增殖抑制率、死亡率均较对照组增加,差异有统计学意义(P<0.01);HBO 2组的细胞增殖抑制率、死亡率较HBO 1组增加,差异有统计学意义(P<0.05);3组SOD的含量比较差异无统计学意义(P>0.05);3组MDA含量比较差异有统计学意义(P<0.01),HBO 1组和HBO 2组人鼻咽癌CNE2Z细胞MDA含量均较对照组增加,差异有统计学意义(P<0.01);HBO 2组的MDA含量较HBO 1组增加,差异有统计学意义(P<0.05).结论 HBO处理增加实验培养的人鼻咽癌CNE2Z细胞增殖抑制率、死亡率,提高鼻咽癌CNE2Z细胞MDA含量,从而提示HBO处理可能是通过增加鼻咽癌CNE2Z细胞MDA含量而抑制鼻咽癌细胞的增殖,促进鼻咽癌细胞的死亡.

목적 연구고압양(HBO)대인비인암세포증식여사망적영향급기궤제.방법 장실험배양적인비인암CNE2Z세포분위대조조、HBO 1조(0.20 MPa)화HBO 2조(0.25 MPa).채용갑기새서기사서(MTT)법분석세포증식억제솔,전화병정(PI)염색관찰세포사망정황,병채용황표령양화매법측정SOD적함량、류대파비타법측정MDA적함량.결과 HBO 1조화HBO 2조적세포증식억제솔、사망솔균교대조조증가,차이유통계학의의(P<0.01);HBO 2조적세포증식억제솔、사망솔교HBO 1조증가,차이유통계학의의(P<0.05);3조SOD적함량비교차이무통계학의의(P>0.05);3조MDA함량비교차이유통계학의의(P<0.01),HBO 1조화HBO 2조인비인암CNE2Z세포MDA함량균교대조조증가,차이유통계학의의(P<0.01);HBO 2조적MDA함량교HBO 1조증가,차이유통계학의의(P<0.05).결론 HBO처리증가실험배양적인비인암CNE2Z세포증식억제솔、사망솔,제고비인암CNE2Z세포MDA함량,종이제시HBO처리가능시통과증가비인암CNE2Z세포MDA함량이억제비인암세포적증식,촉진비인암세포적사망.
Objective To investigate the effect of hyperbaric oxygen (HBO) on the proliferation and death of human nasopharyngeal carcinoma (NPC) ceils and the mechanisms underlying any effect.Methods Cultured CNE2Z line nasopharyngeal carcinoma cells were divided into 3 groups randomly.Group A was a control group;Group B received HBO at 0.20 Mpa;Group C received HBO at 0.25 Mpa.The inhibition ratio of proliferation (IROP) in all groups was detected with an MTT reduction assay,and the mortality rate (MR) was detected by PI staining.Levels of superoxide dismutase (SOD) and malondialdehyde (MDA) were also measured. Results The IROP and MR in the B and C groups increased significantly compared with group A,and there was also a statistically significant difference between groups B and C.There was no significant difference in SOD levels among the groups,but there was a statistically significant difference in the MDA levels,with MDA in groups B and C higher than in group A.There was also a significatnt difference between the B and C groups. Conclusions HBO can increase IROP and MR in human NPC CNE2Z line cells,and elevate their MDA content.This suggests that HBO might inhibit the proliferation of NPC cells and promote their necrosis by increasing MDA levels.
