目的 观察蓝莓对肝纤维化大鼠过氧化物酶体增殖物激活受体γ(PPARγ)和血小板源性生长因子-B(PDGF-B)表达的影响.方法 SD大鼠随机分为对照组、模型组、蓝莓汁预防组(BB组)、丹芍化纤预防组(DSHX组)及蓝莓汁加丹芍化纤预防组(BB+ DSHX组),每组9只.除对照组外,用CCl4复合因素复制大鼠肝纤维化模型,共8周.造模同时,预防组分别予蓝莓汁(1.5 g/100 g)、丹芍化纤(0.1 g/100 g)及蓝莓汁+丹芍化纤(1.5 g/100 g+0.1 g/100 g)每天灌胃1次.8周后分别采用实时荧光定量RT-PCR、免疫组织化学法及Western印迹分析大鼠肝组织Ⅰ型胶原(ColⅠ)、PPARγ及PDGF-B mRNA和蛋白的表达.结果 模型组肝组织PPARγ蛋白的表达明显低于对照组(P<0.05),Col Ⅰ mRNA、蛋白及PDGF-B蛋白的表达均明显高于对照组(P<0.05);各预防组肝组织PPARγ蛋白的表达均明显高于模型组(P<0.05),Col Ⅰ mRNA、蛋白及PDGF-B蛋白的表达均明显低于模型组(P<0.05).结论 蓝莓对大鼠肝纤维化的抑制作用可能与增强PPARγ的活性,从而抑制细胞因子PDGF-B的合成,减少ECM的生成有关.

목적 관찰람매대간섬유화대서과양화물매체증식물격활수체γ(PPARγ)화혈소판원성생장인자-B(PDGF-B)표체적영향.방법 SD대서수궤분위대조조、모형조、람매즙예방조(BB조)、단작화섬예방조(DSHX조)급람매즙가단작화섬예방조(BB+ DSHX조),매조9지.제대조조외,용CCl4복합인소복제대서간섬유화모형,공8주.조모동시,예방조분별여람매즙(1.5 g/100 g)、단작화섬(0.1 g/100 g)급람매즙+단작화섬(1.5 g/100 g+0.1 g/100 g)매천관위1차.8주후분별채용실시형광정량RT-PCR、면역조직화학법급Western인적분석대서간조직Ⅰ형효원(ColⅠ)、PPARγ급PDGF-B mRNA화단백적표체.결과 모형조간조직PPARγ단백적표체명현저우대조조(P<0.05),Col Ⅰ mRNA、단백급PDGF-B단백적표체균명현고우대조조(P<0.05);각예방조간조직PPARγ단백적표체균명현고우모형조(P<0.05),Col Ⅰ mRNA、단백급PDGF-B단백적표체균명현저우모형조(P<0.05).결론 람매대대서간섬유화적억제작용가능여증강PPARγ적활성,종이억제세포인자PDGF-B적합성,감소ECM적생성유관.
Objective To explore the effects of blueberry on the expressions of peroxisome proliferator-activated receptorγ (PPARγ) and platelet-derived growth factor B (PDGF-B) in rat hepatic fibrosis.Methods A total of 45 male Sprague-Dawley rats were randomly divided into control group,CCl4-induced hepatic fibrosis (model group),blueberry prevention (BB group),DSHX prevention (DSHX group) and blueberry + DSHX prevention (BB + DSHX group) (n =9 each).Fibrous liver models of rats were induced by subcutaneous injection of CCl4 and high-lipid/low-protein diet for 8 weeks except for control group.Then the expressions of collagen Ⅰ (Col Ⅰ),PPARγ and PDGF-B were determined by real-time polymerase chain reaction (RT-PCR),immunohistochemistry and Western blot respectively.Results Compared with the control group,the expression of PPARγ decreased and those of Col Ⅰ and PDGF-B were elevated in model group(P < 0.05).The expression of PPARγ increased and Col Ⅰ and PDGF-B decreased in BB,DSHX and BB + DSHX groups as compared to model group (P < 0.05).Conclusion The inhibitory effects of blueberry in CCl4-induced liver fibrosis may be correlated with the activation of PPARγ,the inhibited expression of PDGF-B and the reduced synthesis of extracellular matrix.
