目的 重组腺病毒p53(rAd-p53)对口腔黏膜异常增生细胞(POE-9n,p53-)生长的影响.方法 通过倒置显微镜观察rAd-p53转染前后POE-9n细胞的生长状况及形态变化,并通过流式细胞术及TUNEL荧光染色检测rAd-p53转染后POE-9n细胞凋亡率及其细胞周期分布的改变.结果 经rAd-p53转染72 h后,POE-9n细胞形态和生长方式发生明显变化并发生凋亡,且具有明显的时间依赖性.转染72 h后,POE-9n细胞凋亡最明显,出现明显的细胞周期分布变化,G1期细胞明显增多,S期细胞明显减少,G2期则无明显变化.结论 rAd-p53可以明显抑制POE-9n细胞的增殖,调节细胞周期,使细胞停滞在G1期,并诱导细胞产生凋亡.

Objective To investigate the effect of recombinant adenovirus-p53 (rAd-p53) on growth of oral dysplastic keratinocyte(POE-9n,p53-).Methods Inverted microscope was used to detect the growth status and morphologic changes of POE-9n cell before and after transfection.The cell cycle change and apoptotic rates of POE-9n cell line were analyzed by flow cytometry (FCM) and TUNEL assay.Results After rAd-p53 infection for 72 h,typical morphologic changes were observed in POE-9n cells.By FCM and TUNEL,the time-dependent induced apoptosis was examined.After rAd-p53 infection for 72 h,it was most obvious and induced the arrest cell cycle in G1 stage accompanied with the decrease of S stage cells and no change in G2 stage cells.Conclusions rAd-p53 can strongly inhibit cell proliferation and induce apoptosis and arrest cell cycle in G1 of POE-9n cells.
