目的 探讨苦参碱对肺腺癌细胞凋亡和Bcl-2、Bax蛋白表达的影响.方法 肺腺癌A549细胞进行传代培养后,分别加入30、60、120、240 mg/L的苦参碱,MTT法检测其对A549细胞生长的抑制作用,流式细胞仪检测细胞凋亡及Bcl-2、Bax蛋白的表达.结果 不同浓度的苦参碱对肺腺癌细胞生长均有抑制作用,抑制率与浓度呈正相关.苦参碱可诱导A549细胞凋亡,与作用时间呈正相关.苦参碱能显著降低Bcl-2蛋白表达(P<0.01),而显著升高Bax蛋白表达水平.结论 Bcl-2基因表达降低、Bax基因表达增高可能在苦参碱诱导肺腺癌A549细胞凋亡中起重要作用.

목적 탐토고삼감대폐선암세포조망화Bcl-2、Bax단백표체적영향.방법 폐선암A549세포진행전대배양후,분별가입30、60、120、240 mg/L적고삼감,MTT법검측기대A549세포생장적억제작용,류식세포의검측세포조망급Bcl-2、Bax단백적표체.결과 불동농도적고삼감대폐선암세포생장균유억제작용,억제솔여농도정정상관.고삼감가유도A549세포조망,여작용시간정정상관.고삼감능현저강저Bcl-2단백표체(P<0.01),이현저승고Bax단백표체수평.결론 Bcl-2기인표체강저、Bax기인표체증고가능재고삼감유도폐선암A549세포조망중기중요작용.
Objective To study the expression of Bcl-2 and Bax in the matrine-induced apoptosis of lung adenocarcinoma.Methods After lung adenocarcinoma A549 cells were cultured,matrine of 30,60,120,240 mg/L was added separately.The inhibitory effects of matrine on the growth of A549 cells were tested by MTT method,apoptosis and the expression of Bcl-2 and Bax protein were detected by flow cytometry.Results Matrine with different concentrations had inhibitory effects on the growth of lung adenocarcinoma cells in a concentration-dependent manner.Bcl-2 protein expression was dramatically decreased (P < 0.01) whereas the expression of Bax was increased in A549 cells after treated by matrine.Conclusions The low expression of Bcl-2 and the overexpression of Bax might play an important role in the matrine-induced apoptosis of lung.
