目的 本文通过观察对室尘螨(HDM)过敏的儿童哮喘患者过敏原特异性免疫治疗(allergen specific immunotherapy,SIT)后外周血单个核细胞(peripheral blood mononuclear cells,PBMCs)中IL-10、转化生长因子β1(TGF-β1)表达情况和血清中二者的含量变化,初步探讨儿童过敏性哮喘SIT的机制,从而为哮喘的防治提供新思路.方法 选取郑州大学第一附属医院儿科门诊48例儿童过敏性哮喘患者,分两组,对哮喘组进行标准化SIT 1.5~2年后,采集两组患者的外周静脉血.利用不同的方法检测每组标本PBMCs中IL-10和TGF-β1mRNA以及血清中二者的表达情况,并进行统计学分析.结果 HDM-SIT组中IL-10 mRNA的相对表达量显著升高,是哮喘组的3.29倍,TGF-β1mRNA的相对表达量在HDM-SIT组稍升高,是哮喘组的1.56倍.ELISA检测结果发现与哮喘组比较,HDM-SIT组血清中IL-10和TGF-β1的含量明显升高.结论 本实验进一步证实了哮喘患者SIT有效.在HDM-SIT组IL-10和TGF-β1的表达显著升高,提示二者可能在儿童过敏性哮喘SIT中起重要作用.

Objective The aim of this study was to house dust mite (HDM) children asthma allergen specific immunotherapy (SIT) of allergy of peripheral blood mononuclear cells (PBMCs) changes in IL-10,TGF-β1 expression and the serum content of two,preliminary study on the mechanism of allergic asthma in children with SIT,so as to provide new ideas for prevention and treatment of asthma.Methods The outpatient department of Pediatrics The First Affiliated Hospital of Zhengzhou University in 48 children with allergic asthma patients were divided into two groups,one group was normalized to SIT 1.5-2 years later,peripheral venous blood collected from two groups of patients.Expression of two using different methods for detection of II-10 and TGF-β group of specimens of PBMCs 1 mRNA and serum,and statistical analysis.Results The relative expression level of IL-10 in group HDM-SIT,mRNA increased significantly,is 3.29 times the asthmatic group,the relative expression of TGF-β1 mRNA was slightly increased in HDM-SIT group,is 1.56 times that of asthma group.ELISA test results found that,compared with the asthmatic group,the contents of IL-10 and TGF-β1 HDM-SIT in serum increased significantly.Conclusions This study further confirmed the asthma patients SIT significantly increased in HDM-SIT group; the expression of IL-10 and TGF-β1,TGF-β1,suggesting that IL-10 may play an important role in allergic asthma in children.
