目的 通过观察慢性低氧性肺动脉高压大鼠肺动脉内人第10号染色体缺失的磷酸酶及张力蛋白同源的基因(PTEN)蛋白表达水平的变化,初步探讨PTEN在慢性低氧性肺动脉高压的发生、发展过程中所起的作用.方法 将6周龄健康雄性SD大鼠,随机分为正常对照组、低氧1d、3d、7d、14d和21d组,除对照组外,其他各组先建立慢性低氧肺动脉高压大鼠模型,然后检测各组大鼠右心室收缩压(right ventricle systolic pressure,RVSP)和右心室肥厚指数(right ventricle hypertrophy index,RVHI),采用HE染色观察肺动脉病理学改变,采用Western blot技术检测肺动脉内PTEN蛋白的表达水平.结果 ①与正常对照组(23.76±0.82)mmHg相比,低氧暴露1d、3d、7d、14 d、21d后RVSP均明显上升(P<0.05);RVHI低氧3d、7d、14 d、21d组均较正常对照组(100%)明显上升(P<0.05);低氧暴露3d、7d和21d组肺动脉中膜明显增厚、管腔明显变小.②PTEN和p-PTEN在正常对照组和低氧各组均有表达.低氧各组肺动脉内PTEN蛋白的表达较对照组下降,但差异无统计学意义(P>0.05);而p-PTEN蛋白与PTEN总蛋白表达量的比值随低氧时间的延长有上升趋势,且在慢性低氧21d组(1.71±0.25)较正常对照组(1.00)明显增高(P<0.05).结论 PTEN蛋白表达的降低和p-PTEN蛋白表达的增高可能参与了大鼠慢性低氧性肺动脉高压的发生和发展过程.

목적 통과관찰만성저양성폐동맥고압대서폐동맥내인제10호염색체결실적린산매급장력단백동원적기인(PTEN)단백표체수평적변화,초보탐토PTEN재만성저양성폐동맥고압적발생、발전과정중소기적작용.방법 장6주령건강웅성SD대서,수궤분위정상대조조、저양1d、3d、7d、14d화21d조,제대조조외,기타각조선건립만성저양폐동맥고압대서모형,연후검측각조대서우심실수축압(right ventricle systolic pressure,RVSP)화우심실비후지수(right ventricle hypertrophy index,RVHI),채용HE염색관찰폐동맥병이학개변,채용Western blot기술검측폐동맥내PTEN단백적표체수평.결과 ①여정상대조조(23.76±0.82)mmHg상비,저양폭로1d、3d、7d、14 d、21d후RVSP균명현상승(P<0.05);RVHI저양3d、7d、14 d、21d조균교정상대조조(100%)명현상승(P<0.05);저양폭로3d、7d화21d조폐동맥중막명현증후、관강명현변소.②PTEN화p-PTEN재정상대조조화저양각조균유표체.저양각조폐동맥내PTEN단백적표체교대조조하강,단차이무통계학의의(P>0.05);이p-PTEN단백여PTEN총단백표체량적비치수저양시간적연장유상승추세,차재만성저양21d조(1.71±0.25)교정상대조조(1.00)명현증고(P<0.05).결론 PTEN단백표체적강저화p-PTEN단백표체적증고가능삼여료대서만성저양성폐동맥고압적발생화발전과정.
Objective To observe the changes of PTEN protein expression level in pulmonary arteries of rats induced by hypoxia and preliminarily explores the effect of PTEN in the development of hypoxic pulmonary artery hypertension (HPH).Methods Six week-old healthy male SD rats were randomly divided into normoxia control and normoxia hypoxia for 1,3,7,14 and 21 d groups.Apart from the control group,other groups were exposed to normobaric hypoxia to induce HPH model.Right ventricle systolic pressure (RVSP) and right ventricle hypertrophy index (RVHI) were measured.Pathological changes of the pulmonary arteries were observed by HE staining.PTEN protein expressions in pulmonary arteries were determined by Western blot.Results ① RVSP increased significantly after hypoxic exposure for 1,3,7,14 and 21 d when compared with the normoxia control (23.76± 0.82) mmHg (P <0.05).There were significant rise in RVHI after hypoxic exposure for 3,7,14 and 21 d (P <0.05).And pulmonary arteries was generally thickened and vascular cavity was constricted at 3,7,21 d.② There were expressions of PTEN and p-PTEN in the control and hypoxic groups.PTEN protein expression decreased after chronic hypoxia than normal control groups (P >0.05).There was a tendency of increase in p-PTEN/total PTEN after hypoxia exposure,and the ratio significantly increased at 21 d after hypoxic exposure(1.71±0.25) than that in normal control (1.00)(P <0.05).Conclusions The decrease in protein expression of PTEN and increase of p-PTEN may be involved in the development of the chronic hypoxia-induced pulmonary hypertension.
