目的 研究多氯联苯暴露人群外周血相关基因表达改变.方法 采集32例电子垃圾拆解地居民和48例对照人群外周血样,分离提取总RNA,用实时荧光定量PCR检测CCDC92、CCL22、GZMK、MTDH、STK38L、TMEM175等6个基因的mRNA表达水平.结果 根据双标准曲线法,拆解地居民CCL22基因的表达水平为(4.51±2.37)%,低于对照组的(6.76±4.82) %(t=2.451,P <0.05).根据2-△△Ct法,两者的表达水平分别为(0.54±0.28)倍和(0.81±0.58)倍(t=2.480,P<0.05).根据双标准曲线法,GZMK和MTDH基因的表达水平高于对照组(t=-2.036、-2.228,P<0.05).根据2-△△Ct法,两者的表达水平也高于对照组(t=-2.035、-2.223,P<0.05).两种定量方法存在较好的相关性(r=1.000,P<0.01).结论 CCL22、GZMK和MTDH三个基因在多氯联苯暴露人群中表达异常,可用作多氯联苯暴露人群的效应标志物.

Objective To investigate expression changes of related genes in peripheral blood samples collected from residents exposed to polychlorinated biphenyls (PCBs).Methods Peripheral blood samples were collected from 32 residents in an e-waste recycling site and 48 heaalthy controls.Total RNA was isolated and the expression of CCDC92,CCL22,GZMK,MTDH,STK38L and TMEM175 were detected by quantitative real-time PCR.Results The expression level of CCL22 in the recycling site residents was significantly lower than that of controls by double-standard curves method [(4.51 ± 2.37) %:(6.76 ± 4.82) %,t =2.451,P < 0.05).By 2-△△Ct method,the expression level in two group were (0.54 ± 0.28) times and (0.81 ± 0.58) times (t =2.480,p < 0.05).But the expression levels of GZMK and MTDH were markedly enhanced in exposure group by double-standard curves method (t =-2.036,-2.228,P < 0.05).By 2-△△Ct method,their expression levels were higher than control group (t =-2.035,-2.223,P < 0.05).Good correlations were found between two quantification methods (r =1.000,P < 0.01).Conclusions CCL22,GZMK and MTDH which expressed abnormally in residents in a PCBs pollution site may be used as effect biomarkers for people exposed to PCBs.
