目的 探讨塞来昔布对糖尿病大鼠肾组织胶原纤维、转化生长因子β1(TGFβ1)和Smad7的影响.方法 Wistar大鼠30只,随机分为正常对照组(A组,n=10)、糖尿病组(B组,n=10)和塞来昔布治疗组(C组,n =10).B组和C组于腹腔一次性注射STZ建立糖尿病大鼠模型.模型成立3d后治疗组给予塞来昔布(8 mg/kg·d)进行灌胃,A组和B组用等体积生理盐水灌胃.3个月处死大鼠,肾组织行Masson染色法组织切片,测定胶原纤维面密度水平,RT-PCR测定TGFβ1和Smad7 mRNA表达情况.结果 与正常对照组大鼠相比,糖尿病组大鼠肾组织的胶原纤维、TGFβ1 mRNA表达明显升高(P<0.05),Smad7 mRNA表达明显下降(P<0.05).塞来昔布治疗组大鼠的肾组织胶原纤维、TGFβ1 mRNA表达明显低于糖尿病组,但高于正常对照组(P<0.05);Smad7 mRNA表达高于糖尿病组,但低于正常对照组(P<0.05).结论 塞来昔布可调节糖尿病肾病大鼠肾脏TGFβ1/Smad 7信号通路,减轻肾组织纤维化,延缓糖尿病肾病的发生.

목적 탐토새래석포대당뇨병대서신조직효원섬유、전화생장인자β1(TGFβ1)화Smad7적영향.방법 Wistar대서30지,수궤분위정상대조조(A조,n=10)、당뇨병조(B조,n=10)화새래석포치료조(C조,n =10).B조화C조우복강일차성주사STZ건립당뇨병대서모형.모형성립3d후치료조급여새래석포(8 mg/kg·d)진행관위,A조화B조용등체적생리염수관위.3개월처사대서,신조직행Masson염색법조직절편,측정효원섬유면밀도수평,RT-PCR측정TGFβ1화Smad7 mRNA표체정황.결과 여정상대조조대서상비,당뇨병조대서신조직적효원섬유、TGFβ1 mRNA표체명현승고(P<0.05),Smad7 mRNA표체명현하강(P<0.05).새래석포치료조대서적신조직효원섬유、TGFβ1 mRNA표체명현저우당뇨병조,단고우정상대조조(P<0.05);Smad7 mRNA표체고우당뇨병조,단저우정상대조조(P<0.05).결론 새래석포가조절당뇨병신병대서신장TGFβ1/Smad 7신호통로,감경신조직섬유화,연완당뇨병신병적발생.
Objectives To investigate the effect of celecoxib on diabetic rat kidney tissue collagen,transforming growth factor β1 (TGFβ1) and Smad7.Methods 30 Wistar rats were randomly divided into normal control group (A,n =10),diabetic group (B group,n =10) and celecoxib treatment group (C group,n =10).The diabetic rat models were established in B and C groups by injection of STZ.The Models were established after 3 d treatment group received celecoxib (8mg/kg o d) for gavage,B and C groups with an equal volume of saline.After 3 months,the rats were sacrificed and kidney tissues underwent routine paraffin wax Masson staining tissue sections,the surface density of collagen fibers level was counted.The expression of TGFβ1 and Smad7 mRNA were dectected by RT-PCR.Results Compared with the normal control group,the collagen fibers of kidney tissue in diabetic rats,TGFβ1 Smad7 mRNA expression was significantly increased(P <0.05),Smad7 mRNA expression was significantly decreased(P <0.05).The collagen fibers in Celecoxib treated rats,TGFβ1 mRNA expression was significantly lower than the diabetic group and higher than the normal control group (P < 0.05),Smad7 mRNA expression was increased than the diabetic group and decreased than the normal control group(P < 0.05).Conclusions Celecoxib can alleviate development of renal fibrosis in rats.The possible mechanism might attribute to the regulation of collagen fiber,transforming growth factor-β1 and Smad7.
