目的 检测良性前列腺增生、前列腺癌标本中自噬相关基因Beclin-1、LC3的表达及其与凋亡基因P53、Bcl-2的关系,并探讨其意义.方法 分别取34例前列腺癌组织和50例良性前列腺增生组织(术前均未服5α-还原酶抑制剂),采用Western blot方法检测其Beclin-1和LC3的表达.再将96例良性前列腺增生病理标本分为服药组(服用5α-还原酶抑制剂,共计55例)、对照组(未服用任何药物,共计41例),采用免疫组化方法检测Beclinl、LC3、P53和Bcl-2的表达.结果 Western blot检测Beclin-1、LC3在良性前列腺增生中的表达比前列腺癌高(P<0.001),不同Gleason分级间前列腺癌的Beclin-1、LC3表达未见差别(P>0.05).免疫组化检测Beclin-1、LC3在服药组中的表达增高,而Bcl-2、P53降低(P<0.05);LC3与Beclin-1呈正相关(r=0.345,P=0.001),与BCL-2呈负相关(r=-0.216,P=0.035);Beclin-1与BCL-2呈负相关(r=-0.402,P≤<0.001).结论 自噬相关基因Beclin-1、LC3参与前列腺癌的发生发展;而服用5α-还原酶抑制剂的良性前列腺增生患者,因体内雄激素降低而诱导的自噬相关基因Beclin-1、LC3表达增强,并伴随Bcl-2和P53降低,这可能使前列腺癌变的风险降低.

Objectives To investigate the expression of autophagy-related genes Beclin-1 、LC3 in benign prostatic hyperplasia tissue and prostate cancer tissue,and to explore the correlations between it and apoptosis-related gene P53 、Bcl-2.Methods Tested using western blotting to confirm the expressions of Beclinl,LC3 in 34 cases of BPH and 50 cases of prostate cancer.Then,Immunohistochemistry were used to detect the expressions of Beclinl,LC3,P53 and Bcl-2 in 55 cases of BPH who have taken medicine and in the other 41 cases who have not.Results We found that Beclin-1,LC3 expression were higher in BPH tissues than in Prostate cancer tissues(P <0.001),and have no statistics difference in different Gleason stage of prostate cancer(P > 0.05).Then,IHC showed that Beclin-1,LC3 expression were significantly higher in taken-medicine cases,however,Bcl-2 and were lower (P < 0.05).LC3 expression was found to have positive correlation with Beclin-1 (r =0.345,P =0.001) and to have negative correlation with BCL-2 (r =-0.216,P =0.035).In addition,Beclin-1 have positive correlation with BCL-2 (r =-0.402,P ≤0.001).Conclusions Autophagy-related genes,Beclin-1 and LC3,might be involved in the genesis and development of Pca.In addition,it was higher when patients with BPH had taken 5α -reductase inhibitor due to androgen reduction.As a result,autophagy induced by that might reduce the occurrence of Pca risk.
