目的 探讨人类免疫缺陷病毒感染者包膜蛋白V4区变异与CD4+ T细胞数之间的关系.方法 本研究从Los Alamos HIV序列数据库中选取369条HIV-1 B亚型且已知CD4+ T细胞数的V4区氨基酸序列.分析CD4+ T细胞数与V4区氨基酸序列长度、潜在糖基化位点(PNGS)及净电荷数的相关性.V4区氨基酸序列按CD4+ T细胞数高低分成两组:CD4+ T细胞数< 200个/μL组(n=216)和-CD4+ T细胞数≥200个/μL组(n=153),比较两组V4区氨基酸的差异.结果 CD4+ T细胞数与V4区氨基酸序列长度及PNGS数正相关(r=0.248,0.136,P均<0.01),而与V4区净电荷数负相关(r=-0.154,P<0.01).CD4+ T细胞数<200个/μL与CD4+ T细胞数> 200个/μL组相比,V4区氨基酸序列长度较短(P<0.01)、PNGS数较少(P<0.05).但CD4+ T细胞数高低两组V4区净电荷数之间无统计学意义(P=0.17).结论 V4区变异和HIV-1感染的病程具有相关性,随着CD4+ T细胞数的降低,V4区序列变短、PNGS数减少.

Objective To characterize the correlation between the variation of V4 sequence and CD4+ T cell counts.Methods 369 V4 sequences with CD4+ T cell counts known were downloaded from the Los Alamos HIV Sequence Database.Correlations between CD4+ T cell counts and length,the number of PNGS,net charge were assessed for univariate comparisons.Furthermore,All the sequences were classified into high CD4+ T cell counts (≥200/μL) group and low CD4+ T cell counts group (< 200/μL).No-parametric test was used to discover the different between the two groups.Results There were significant positive correlations between the CD4+ T cell counts and length,PNGS(r =0.248,0.136,P < 0.01).However,the results showed a negative association between the CD4+ T cell counts and net charge(r =-0.154,P <0.01).Moreover,there were less number of amino acids (P < 0.01) and PNGS (P < 0.05) of V4 sequences with CD4+ T cell counts < 200 cells/μL.Conclusion The length and PNGS of V4 were decreased with the decline of CD4+ T cell counts,which indicated that there was a correlation between the variation of V4 and CD4+ T cell counts.
