背景 血管的通透性主要由跨细胞途径和细胞旁途径两部分组成.在炎症反应过程中,细胞旁途径是增加血管通透性的主要因素.紧密连接(tight junction,TJ)是细胞旁途径的重要组成部分,破坏它将导致细胞通透性的增加. 目的 分析TJ的信号机制,更好地理解其保护血管屏障的功能. 内容 对影响TJ的几个重要的信号通路进行综述,揭示其影响细胞通透性的信息传递和调控作用. 趋向 TJ的完整性可以抑制水肿液的产生,今后将进一步探索其在炎症性疾病中的具体应用,从而更好地诊断疾病,预测疾病的发生发展及疗效.

Background Vascular permeability is primarily composed of transcellular and paracellular pathways,with the latter being a major contributor to the increasing of vascular permeability in the process of inflammation.Tight junction (TJ) is the important component of paracellular pathways.The destruction of TJ will induce the increasing of vascular permeability.Objective This article is to analyze signaling mechanisms of TJ and to explain its function of protecting blood vessels barriers.Content This article reviewed several important signaling mechanisms which affect the TJ,revealed its information transfer and regulation of cell permeability.Trend The integrity of TJ can restrain the generation of edema fluid and future research will focus on its specific application in inflammatory disease,in order to diagnose better and predict development and therapeutic effect of disease.
