目的 探讨阿霉素(ADM)干预下人肾上腺皮质癌裸鼠模型干细胞标志CD133、ABCG2的表达及意义.方法 SW-13细胞系建立裸鼠皮下移植瘤模型,ADM体内干预后原代培养获得ADM组细胞株,未予干预直接原代培养获得对照组细胞株.用单细胞成克隆技术检测细胞的增殖能力,免疫组化法和流式细胞法检测肿瘤干细胞标志物CD133、ABCG2的表达.结果 ADM组细胞株的克隆形成率高于对照组(P<0.05);流式细胞法和免疫组化法提示ABCG2在两组细胞中均有阳性表达,ADM组细胞株的流式细胞表达率高于对照组细胞株(P<0.05);以上两种方法均表明CD133在两组中阴性表达.结论 ADM组细胞具有一定的肿瘤干细胞特征,ABCG2参与肾上腺皮质癌细胞耐药机制,可能为分子靶向治疗的潜在靶点.
目的 探討阿黴素(ADM)榦預下人腎上腺皮質癌裸鼠模型榦細胞標誌CD133、ABCG2的錶達及意義.方法 SW-13細胞繫建立裸鼠皮下移植瘤模型,ADM體內榦預後原代培養穫得ADM組細胞株,未予榦預直接原代培養穫得對照組細胞株.用單細胞成剋隆技術檢測細胞的增殖能力,免疫組化法和流式細胞法檢測腫瘤榦細胞標誌物CD133、ABCG2的錶達.結果 ADM組細胞株的剋隆形成率高于對照組(P<0.05);流式細胞法和免疫組化法提示ABCG2在兩組細胞中均有暘性錶達,ADM組細胞株的流式細胞錶達率高于對照組細胞株(P<0.05);以上兩種方法均錶明CD133在兩組中陰性錶達.結論 ADM組細胞具有一定的腫瘤榦細胞特徵,ABCG2參與腎上腺皮質癌細胞耐藥機製,可能為分子靶嚮治療的潛在靶點.

Objective To explore the expression of CD133 and ABCG2 in adrenocortical carcinoma (ACC) xenograft treated by adriamycin in vivo,and to preliminarily analyze the significance.Methods Established the nude mice model of SW-13 transplanted subcutaneously,one group was intervened with ADM in vivo and obtained ADM group cells by primary culture,the other group was not intervened with ADM and obtained control group cells by primary culture.Cell proliferation capacity was examined by single-cell clone formation assay.The expression of CD133 and ABCG2 were detected by flow cytometry and immunohistochemistry.Results The colony formation rate of ADM group was stronger than that of control group (P <0.05).Flow cytometry and immunohistochemistry showed that ABCG2 were positive in both groups,the expression rate of ABCG2 by flow cytometry was higher in ADM group(P < 0.05),but the two methods indicated CD133 was negative in both groups.Conclusions The ADM group cells have certain characteristics of cancer stem cells.ABCG2 is associated with the formation of drug resistance in ACC and may be a potential therapeutic target.
