目的 评价不同剂量血红蛋白类携氧载体(HBOC)对大鼠肺动脉平滑肌细胞大电导钙离子激活钾离子通道(BKCa)电流的影响.方法 采用二步酶消化法急性分离Sprague Dawley(SD)大鼠肺动脉平滑肌细胞.BKCa电流采用四氨基吡啶(4-AP)进行分离并使用全细胞电压钳记录.选取急性分离后状态良好的大鼠肺动脉平滑肌细胞30个,根据计算机生成的随机数将其分为5组,每组6个平滑肌细胞,分别给予血红蛋白含量(kg/L)为0.25×10-2,0.50×10-2,1.00×10-2,2.00×10-2,3.00×10-2的HBOC处理.观察不同剂量HBOC对BKCa电流的影响,并与给予HBOC前的BKCa电流相比较(实验过程中对动物的处置符合动物伦理学标准).结果 经血红蛋白含量为0.25×10-2 kg/L的HBOC处理后,BKCa电流轻度增加,BKCa电流值与基础值比较,增加3.90%;分别给予血红蛋白含量(kg/L)为0.50×10-2,1.00×10-2,2.00×10-2,3.00×10-2的HBOC处理后,BKCa电流呈不同程度下降,BKCa电流值与基础值比较,分别下降8.04%,28.61%,46.53%和65.97%,HBOC对BKCa电流的抑制作用呈剂量依赖性(b=0.931 6,P=0.007).结论 HBOC呈剂量依赖性抑制大鼠肺动脉平滑肌细胞BKCa电流.BKCa电流的下降可能是HBOC导致血管收缩的原因.

Objective To evaluate the effect of different doses of hemoglobin-based oxygen carriers (HBOC) on the large conductance calcium-activated potassium channels (BKCa) currents of rat pulmonary artery smooth muscle cells.Methods Vascular smooth muscle cells were acutely isolated from rat pulmonary arteries with two steps of enzymatic digestion.BKCa currents were separated with 4-aminopynidine (4-AP),and the effects of HBOC on BKCa currents were evaluated by the whole-cell patch-clamp technique.Thirty rat pulmonary artery smooth muscle cells which were in good condition were selected into this study.They were divided into five groups according to the computer-generated random numbers and six cells each group.The cells in each group were randomly given one of five HBOC concentrations (kg/L)which were 0.25 ×10-2,0.50×10-2,1.00×10-2,2.00×10 2,3.00×10-2.The effects of HBOC on amplitude of BKCa currents were recorded and were compared with the BKCa currents before giving HBOC.All experiments were carried out in accordance with relevant regulatory standards for animal ethics.Results We find that acute addition of 0.25 × 10-2 kg/L HBOC into the bath solution could make the BKCa currents increased 3.90%.While the HBOC concentrations (kg/L) were 0.50× 10-2,1.00× 10-2,2.00×10 2 and 3.00 × 10-2,the BKCa currents were significantly inhibited in a dose-dependent manner (b =0.931 6,P =0.007),with inhibition of 8.04%,28.61%,46.53% and 65.97% respectively.Conclusions HBOC could inhibit the BKCa currents of rat pulmonary artery smooth muscle cells in a dosedependent manner.And the inhibition may contribute to HBOC-induced vasoconstriction.
