目的 了解1株多耐药肺炎克雷伯菌JM45株对氨基糖苷类药物耐药的遗传学背景.方法 采用罗氏454高通量测序技术对肺炎克雷伯菌JM45株做全基因组测序(完成图),再分析氨基糖苷类修饰酶基因、16S核糖体RNA(rRNA)甲基化酶基因检测状况,并和其他已经完成全基因组测序的革兰阴性菌作比较基因组学分析.结果 最终得到肺炎克雷伯菌JM45株1条完整的基因组(染色体)序列及2条质粒序列.基因组(染色体)序列为5 273 812 bp(GC含量65.8%),质粒1序列为317 156 bp(GC含量53.0%),质粒2序列为12 209 bp(GC含量55.3%).在质粒1中发现了氨基糖苷类腺苷转移酶基因aadA2(KPN_5186)和16S rRNA甲基化酶基因rmtB(KPN_5243).由全基因组测序可见aadA2基因位于整合子中,排序为int ⅠA-dfrA12 aadA2 orf3/qacE;rmtB基因位于转座子中,排序为tnpA-TEM1 rmtB.结论 携带aadA2和rmtB基因是JM45株耐氨基糖苷类药物的主要原因.
目的 瞭解1株多耐藥肺炎剋雷伯菌JM45株對氨基糖苷類藥物耐藥的遺傳學揹景.方法 採用囉氏454高通量測序技術對肺炎剋雷伯菌JM45株做全基因組測序(完成圖),再分析氨基糖苷類脩飾酶基因、16S覈糖體RNA(rRNA)甲基化酶基因檢測狀況,併和其他已經完成全基因組測序的革蘭陰性菌作比較基因組學分析.結果 最終得到肺炎剋雷伯菌JM45株1條完整的基因組(染色體)序列及2條質粒序列.基因組(染色體)序列為5 273 812 bp(GC含量65.8%),質粒1序列為317 156 bp(GC含量53.0%),質粒2序列為12 209 bp(GC含量55.3%).在質粒1中髮現瞭氨基糖苷類腺苷轉移酶基因aadA2(KPN_5186)和16S rRNA甲基化酶基因rmtB(KPN_5243).由全基因組測序可見aadA2基因位于整閤子中,排序為int ⅠA-dfrA12 aadA2 orf3/qacE;rmtB基因位于轉座子中,排序為tnpA-TEM1 rmtB.結論 攜帶aadA2和rmtB基因是JM45株耐氨基糖苷類藥物的主要原因.

Objective To investigate genetic background of aminoglycoside resistance in multi drug resistant Klebsiella pneumoniae JM45 isolate.Methods Whole genome sequencing (complete graph) was performed by high throughput Roche 454 sequencing approach.Aminoglycoside modifying enzyme genes and 16S rRNA methylase genes were further analyzed,and comparative genomics analysis was performed with other Gram-negative bacteria.Results A complete genome (chromosome) sequence and two plasmid sequences were obtained in JM45.The size of chromosome was5 273 812 bp (GC content:65.8%),the size of plasmid 1 was 317 156 bp (GC content:53.0%),and that of plasmid 2 was 12 209 bp (GC content:55.3%).Aminoglycoside adenosine transferase gene (aadA2,Feature ID:KPN_5186) and 16S rRNA methylase genes (rmtB,Feature ID:KPN_5243) were positive in plasmid 1.According to whole genome sequencing,aadA2 located in the integron and the order was intⅠA-dfrA12 aadA2 orf3/qacE; rmtB located in the transposon and the order was tnpATEM-1-rmtB.Conclusion aadA2 and rmtB play a key role in aminoglycoside resistance in JM45.
