目的 评价超声辐照载血管内皮细胞生长因子抑制剂(vascular endothelial growth factor inhibitor,VEGFI)脂质微泡对结肠癌的治疗效果.方法 将稳定表达绿色荧光蛋白的结肠癌细胞系接种于Balb/c裸鼠皮下,建立结肠癌移植瘤模型.将48只皮下种植结肠癌移植瘤的Balb/c裸鼠随机分为4组:A组(12只),空白对照组,不接受超声造影及辐照;B组(12只),接受裸脂质微泡超声造影及假照;C组(12只),裸脂质微泡超声造影及超声辐照;D组(12只),载VFGFI脂质微泡及超声辐照.辐照前和3次辐照后第1d、第3d、第7d、第11d、第16d分别对各组裸鼠测量体质量和荧光摄片,测量肿瘤大小,计算肿瘤体积,绘制肿瘤体积生长曲线及裸鼠体质量生长曲线.16d后处死裸鼠,切除并分离肿瘤组织,测量各组肿瘤质量.结果 辐照前各组裸鼠体质量和肿瘤体积差异无统计学意义(P>0.05);随访期间,A组与B组肿瘤体积持续增大,两组间比较差异无统计学意义(P>0.05);辐照后第7d,C组及D组肿瘤体积均小于A组,差异有统计学意义(P<0.01),C组与D组之间比较差异无统计学意义(P>0.05);第7d后C组肿瘤体积变化不明显,到第16d肿瘤体积仍然小于A组,差异有统计学意义(P<0.01),而D组肿瘤体积进一步缩小,与C组比较差异有统计学意义(P<0.05).切除肿瘤组织,D组肿瘤质量低于C组(P<0.05),C组低于A组(P<0.05),而A、B组之间肿瘤质量差异无统计学意义(P>0.05).观察期间各组裸鼠体质量差异无统计学意义(P>0.05).结论 载VEGFI微泡联合超声辐照可以增强对结肠癌的治疗效果.

Objective To estimate the therapeutic efficacy of colon cancer with ultrasound destruction of vascular endothelial growth factor inhibitor(VEGFI)-carrying lipid microbubbles.Methods Colon tumor xenografts cells stably expressing green fluorescent protein were inoculated subcutaneously in 48 Balb/c nude mice and tumor (5-20 mm) developed.The mice were randomly divided into four groups:Group A (12 mice),as controls,without any treatment.Group B (12 mice) underwent contrast-enhanced ultrasound (CEUS) examination and sham ultrasound radiation.Group C (12 mice) were given lipid microbubbles combined with ultrasound radiation.Group D (12 mice) underwent VEGFI-carrying lipid microbubbles injection combined with ultrasound radiation.Fluorescence imaging was performed and nude mice body weight was measured respectively before and 1 day,3 days,7 days,11 days,16 days after three times ultrasound radiation.Tumor volume was calculated,tumor growth curve and the nude mice body weight growth curve were drawn.The mice were sacrificed and tumor specimens were weighed.Results There was no significant difference in the body weight of nude mice and tumor volume before radiation among the four groups (P > 0.05).During the follow-up period tumor continued to increase in volume in group A and group B,and the difference between the two group was not statistically significant (P >0.05).Compared with the control group,tumor volumes in group C and group D were smaller 7th day after ultrasound radiation,the difference was statistically significant (P <0.01),while there was no significant difference in group C and group D (P >0.05).There was no significant change in tumor volume in group C after 7 days,yet tumor volume was still smaller than that in group A after 16 days,the difference was statistically significant (P <0.01).Compared with group C,tumor volume in group D was further reduced,the difference was statistically significant after 16 days (P <0.01).When tumor tissue was removed,tumor weight in group C was lower than that in group A and group B (P <0.05),while tumor weigh in group D was significantly lower than that in any other group (P <0.05).There was no significant difference in tumor weight between group A and group B (P > 0.05).During the observation period there was no significant difference in the body weight of nude mice among the four groups (P >0.05).Conclusions Ultrasound destruction of VEGFI-carrying lipid microbubbles can improve the therapeutic efficacy of colon cancer.
