目的 自制骨生长定量检测盒检测人工成骨材料的成骨效能,探讨骨生长定量检测动物模型的可行性.方法 取贵州小型猪8头,将自制的可重复使用的骨生长定量检测盒植入四肢骨骼中.第1次手术植入骨生长定量检测盒建模,骨生长定量检测盒双腔内芯均不填充任何材料;第2次手术更换骨生长定量检测盒内芯,一侧腔室填充重组合异种骨为实验组,另一侧腔室空置为对照组;第3次手术更换骨生长定量检测盒内芯,两侧腔室填充同第2次手术;第4次手术处死动物,取出骨生长定量检测盒.每次手术间隔12周.对第3、4次手术取出的骨生长定量检测盒内芯中的实验组及对照组标本行组织学检测(新生骨骨小梁宽度和骨小梁面积百分数)与生物力学检测(抗压极限强度及杨氏模量).结果 实验组内芯有灰白色组织生长,镜下可见骨组织形成;对照组空腔内有褐色组织生长,镜下仅有纤维组织生成.实验组骨小梁宽度为(128.15±39.96)mm、骨小梁面积百分数为31.38%±2.67%,与对照组的差异有统计学意义.不同动物个体间、前肢与后肢、左侧肢与右侧肢的骨小梁宽度、骨小梁面积百分数、抗压极限强度及杨氏模量的差异均无统计学意义.结论 骨生长定量检测盒内的人工成骨材料具有成骨诱导作用;可重复使用的骨生长定量检测盒动物模型可用于人工成骨材料诱导成骨效能的定量检测;动物模型的不同个体及不同实验部位对人工成骨材料的诱导成骨效能无明显影响.

Objective To investigate the feasibility of bone harvest chamber on quantitative detection of the bone performance efficacy of artificial osteoinductive materials in animal model.Methods Repeated bone harvest chamber was implanted into bones of Guizhou mini-pigs (n=8).In the first surgery,nothing was filled in the inner cores of the chambers.After 12 weeks,the inner cores of the chambers were exchanged at the second surgery.In the experimental group,artificial osteoinductive materials were filled in one core,nothing filled in the other core was as control.After 12 weeks,the inner cores were exchanged again.Finally,the bone harvest chambers were removed after 12 weeks.The inner core samples were randomly divided into two groups.The thickness of new trabeculae and areas percentage were measured by pathological analysis in one group.In the other group,the ultimate compressive strength and young modulus were surveyed by biomechanical test.Results The hoar tissues were observed by naked eye in experimental group,but only brown tissues were observed in control group.While the new bone tissues were observed in experimental group,only fibrous tissues were observed in control group.The thickness of new trabeculae was 128.15±39.96 mm and the areas percentage of new trabeculae was 31.38%±2.67% in experimental group.The results of histological analysis and biomechanical tests were significantly different between experimental group and control group.The results confirmed that artificial osteoinductive materials could induce the formation of new bone.There were no significant differences within different individuals and different experimental limbs of Guizhou mini-pigs on the thickness of new trabeculae,areas percentage,the ultimate compressive strength and young modulus in experimental group.Conclusion Artificial osteoinductive materials can induce bone formation in inner core of repeated bone harvest chamber.Repeated bone harvest chamber can be used to quantitatively detect the bone formation of artificial bone materials.There are no significant difference in bone formation efficacy of artificial osteoinductive materials within different individuals and experimental limbs of Guizhou mini-pigs.
