目的 观察HBV对原蛋白转化酶Furin表达的影响,明确HBV在宿主细胞内适应生存过程中宿主细胞内Furin表达的变化,为今后探索提高抗病毒治疗疗效提供理论基础.方法 采用荧光定量PCR、Western blot及免疫细胞化学技术比较慢性乙型肝炎肝组织和正常肝组织之间,以及稳定表达HBV复制的HepG2.2.15细胞株和前体细胞HepG2之间的Furin表达变化,从mRNA、蛋白水平、组织水平观察HBV对肝细胞内Furin表达的影响.Furin mRNA及蛋白表达结果根据3次独立试验的倍数均数和标准差进行t检验. 结果 在慢性乙型肝炎肝组织中Furin表达下调,在HepG2.2.15细胞上,FurinmRNA(与HepG2细胞比较为下调50.1%±4.8%对比1)及蛋白表达水平均被下调,P值均< 0.05.结论 HBV通过下调宿主细胞内Furin可能有利于病毒自身复制.

Objective To study the effects of hepatitis B virus (HBV) infection on the expression of Furin,an important proprotein convertase,in liver cells to provide insights towards its potential as a therapeutic target for improved antiviral efficacy.Methods Furin expression was measured in human liver specimens (infected tissues from patients with chronic HBV hepatitis vs.normal tissues from healthy donors) and in hepatoma cell lines (HBV-infected HepG2.2.15 cells vs.uninfected parental cell lines HepG2) using quantitative real-time RT-PCR (for mRNA),western blotting and immunohistochemistry (for protein).Results Compared to the uninfected tissues and cells,the HBV-infected tissue and cells showed down-regulated expression of furin at both the mRNA and protein levels.In particular,the HepG2.2.15 cells showed ~50% less furin mRNA expression than the HepG2 cells and the difference was statistically significant (P < 0.05).Conclusion HBV may suppress the host cell's expression of furin,possibly to benefit its survival and replication in the host cell.
