目的 探讨高压氧(HBO)对小鼠结肠癌移植瘤生长的抑制作用及对肿瘤组织血管内皮细胞生长因子(VEGF)表达和肿瘤微血管密度(MVD)的影响.方法 建立小鼠结肠腺癌细胞CT26移植瘤模型16只,按数字表法随机分为对照组(8只)和HBO组(8只).HBO组每天HBO暴露1次,每次60 min,共15次.HBO处理结束后检测肿瘤生长情况,计算抑瘤率,并采用免疫组织化学法检测移植瘤组织VEGF表达和肿瘤MVD的改变.结果 与对照组[(2.692±0.391)cm3]比较,HBO组[(1.729±0.402)cm3]肿瘤的增长速率减慢(P<0.05),且HBO组VEGF的表达(1.82±0.43)及MVD计数(23.43±4.05)与模型对照组VEGE(2.37±0.16)和MVD(31.57±6.32)比较均显著性降低(P<0.05).结论 HBO可能通过抑制VEGF的表达、降低肿瘤血管生成进而抑制肿瘤的生长.

Objective To investigate the effect of hyperbaric oxygen on tumor growth and the expression of vascular endothelial growth factor(VEGF)and microvessel density(MVD)of colon cancer in mice.Methods CT26 transplantation tumor of colonic carcinoma cell mouse model was established,and the mice were randomly divided into the blank control group,the model control group and the hyperbaric oxygen(HBO)group.The HBO group was exposed to hyperbaric oxygen once a day,60 min a session,with a total of 15 sessions.Following HBO treatment,tumor growth was detected,inhibitory rate of tumor growth was calculated and the expression of VEGF of transplantation tumor tissue and changes in MVD were detected by using immunohistochemistry.Results When compared with the control group[(2.692±0.391)cm3],the tumor growth rate for the animals in the HBO group[(1.729±0.402)cm3]decreased(P<0.05),and the expression of VEGF and MVD in the animals of the HBO group(2.37±0.16,31.57±6.32)all decreased significantly(P<0.05).Conclusions HBO might inhibit tumor growth through the access of inhibiting the expression of VEGF and decreasing tumor angiogenesis.
