目的 检测高压氧(hyperbaric oxygen,HBO)对实验兔颈内动脉和基底动脉内皮素-1(ET-1)及内皮型一氧化氮合酶(eNOS) mRNA表达的影响,探讨HBO暴露致脑血管前循环收缩、后循环扩张的可能机制.方法 健康成年新西兰白兔24只,按数字表法随机分为实验组(n=12)和对照组(n=12).实验组每日持续暴露于0.22 MPa HBO下60 min,连续3d;对照组正常饲养,不予任何处理.实时荧光定量PCR法(FQ-RT-PCR)检测ET-1、eNOS mRNA在2组颈内动脉和基底动脉的表达水平.结果 ET-1、eNOS mRNA在实验兔颈内动脉和基底动脉均有微量表达.与对照组相比,HBO可使实验组颈内动脉ET-1 mRNA表达明显增高[(1.679 ±0.097) vs (1.000±0.017)],eNOS mRNA表达明显下降[(0.571 ±0.039) vs (1.000 ±0.023)];基底动脉ET-1 mRNA表达明显下降[(0.565 ±0.069) vs(1.000±0.034)],eNOS mRNA表达明显增高[(1.667 ±0.134) vs(1.000±0.014)],差异均有统计学意义(P<0.01).结论 HBO可通过调节ET-1、eNOS mRNA在颈内动脉和基底动脉的相对表达量而发挥收缩前循环、扩张后循环的作用.

Objective To detect the effects of hyperbaric oxygen (HBO) on the expressions of endothelin-1 (ET-1) and endothelial nitric oxide synthase (eNOS) mRNA in the internal carotid artery (ICA) and basal artery (BA) of rabbits,and also to explore the mechanism involved in the constriction of the cerebrovascular anterior circulation and dilation of the cerebrovascular posterior circulation.Methods Twenty-four healthy adult New Zealand rabbits were randomly divided into 2 groups:the experimental group and the control group,each consisting of 12 animals.The rabbits in the experimental group were exposed to HB0 at 2 ATA for 60 minutes a day for 3 successive days,while the rabbits in the control group were normally fed without any treatment.Real-time-FQ-PCR was used to detect expression levels of ET-1 and eNOS mRNA in both ICA and BA of the 2 groups.Results Slight expressions of ET-1 and eNOS mRNA could be detected in ICA and BA of the experimental group.Significant increase in the expression of ET-1 mRNA and marked decrease in the expression of eNOS mRNA in ICA could be detected in the experimental group (1.679 ± 0.097),as compared with that of the control group (1.000 ± 0.017).At the same time,HBO exposure could significantly induce the expression of ET-1 mRNA and marked increase in the expression of eNOS mRNA in BA,with statistical significance (P < 0.01).Conclusions Through regulation of the expression amount of ET-1 and eNOS mRNA in both ICA and BA,HBO could play a role in the contraction of cerebrovascular anterior circulation and dilation of cerebrovascular posterior circulation.
