目的 研究高压氧对脑出血大鼠脑内血管新生因子的影响及作用机制.方法 将60只大鼠按数字表法随机分为假手术组、脑出血组和高压氧组,每组20只大鼠.观察各组大鼠行为学改变情况;检测各组大鼠脑内增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)、Ⅷ因子相关抗原(von Willebrand factor,vWF)、缺氧诱导因子-1(hypoxia inducible factor-1,HIF-1)、血管内皮生长因子(vascular endothelial growth factor,VEGF)的蛋白表达量.结果 实验大鼠行为学评分,高压氧组与脑出血组比较14、21、28 d差异均有统计学意义(P<0.01);血肿周围脑组织的HIF1-α蛋白表达均高于脑出血组,2组14、21、28 d比较差异有统计学意义(P<0.01);血肿周围脑组织的PCNA蛋白表达高压氧组均高于脑出血组,7、14、21、28 d差异均有统计学意义(P<0.05);VEGF蛋白表达高压氧组高于脑出血组,2组14、21、28 d比较差异均有统计学意义(P<0.01);高压氧组vWF蛋白表达均高于脑出血组,2组7、14、21、28 d比较差异均有统计学意义(P<0.01).结论 高压氧能通过促进血管新生相关因子HIF1-α、VEGF、PCNA及vWF蛋白的表达,促进脑出血损伤组织血管的新生,增加血肿吸收,促进行为能力的恢复和机体的康复.

Objective To investigate the effects of hyperbaric oxygen (HBO) on intracephalic angiogenesis in rats with intracerebral hemorrhage (ICH) and the mechanism involved.Methods The healthy SD rats were randomly divided into the sham surgery group (group A),the intracerebral hemorrhage group (group B) and the HBO group (group C).Observations were made on changes in rat behavior of each group.Expression levels of proliferating cell nuclear antigen (PCNA),yon Willebrand 's factor (vWF),hypoxia inducible factor-1 (HIF-1 α) and vascular endothelial growth factor (VEGF) were detected in the brain of rats of all the groups.Results Significant differences could be noted in behavior scores of the HBO group at d 14,d 21 and d 28,as compared with those of group B,with obvious statistical significance (P < 0.01).The expression of HIF1-α in the brain tissue around the hematoma was higher than that of group B,and statistical significance could be seen in the expression levels at d 14,d 21 and d 28,when comparisons were made between the 2 groups (P < 0.01).The expression of PCNA in the brain tissue around the hematoma for the animals of the HBO group was higher than that of group B,and statistical significance could be seen in the expression levels at d 14,d 21 and d 28,when comparisons were made between the 2 groups(P < 0.05).The expression of VEGF for the HBO group was higher than that of group B,and statistical significance could be seen in the expression levels at d 14,d 21 and d 28,when comparisons were made between the 2 groups(P <0.01).The expression of vWF for the HBO group was also higher than that of group B,and statistical significance could also be seen in the expression levels at d 14,d 21 and d 28 (P < 0.01).Conclusions HBO could accelerate angiogenesis of the damaged tissue induced by cerebral hemorrhage,promote absorption of hematoma,enable recovery of behavioral capacity and enhance rapid rehabilitation of the body,through expressions of such related factors as HIF-1 a,VEGF,PCNA and vWF proteins.
