目的 应用流式细胞仪测定小檗碱对白色念珠菌细胞周期的影响,初步探索小檗碱抑制白色念珠菌的作用机制.方法 将白色念珠菌(ATCC76615)培养在含不同质量浓度(0.16、0.08、0.04及0 g/L)小檗碱的酵母浸出粉胨葡萄糖培养基中,培养6、12和24h后应用流式细胞仪检测该药物对白色念珠菌(ATCC76615)细胞生长周期的影响.结果 白色念珠菌(ATCC76615)在与小檗碱作用12h时,随着药物质量浓度的增高(0、0.04、0.08、0.16 g/L)增殖指数(分别为98.8、82.4、75.6、54.1)均显著降低(P<0.001).结论 小檗碱对白色念珠菌的细胞生长周期具有抑制作用,其最佳作用时间为12h.

Objective To determine the effect of berberine on cell cycle of Candida albicans.Methods Flow cytometry (FCM) was used to determine the effect of berberine on cell cycle of Candida albicans.After Candida albicans (ATCC76615) was incubated in yeast extract peptone dextrose (YPD)medium which contained different concentration of berberine for 6 hours,12 hours and 48 hours respectively,the cell cycle was detected by FCM.The proliferation index(PI) was calculated.Results PI was significantly inhibited when Candida albicans (ATCC76615)was incubated with berberine for 12 hours (0 g/L:98.8,0.04 g/L:82.4,0.08 g/L:75.6,0.16 g/L:54.1).Conclusions The berberine could inhibit the cell cycle of Candida albicans in vitro,wbich may be the potential mechanism of antifungal effect of berberine against Candida albicans.
