目的 观察4.0代聚酰胺-胺对敏感牙本质小管的生物矿化作用,以期为聚酰胺-胺在牙本质敏感治疗中的应用奠定实验基础.方法 收集完整离体第三磨牙36颗,切取釉质牙骨质界处约2 mm厚牙本质,制备敏感牙本质样本,将其一分为二,利用随机数字表法随机选择1/2作为对照组,另1/2作为实验组.对照组用去离子水处理,实验组用4.0代聚酰胺-胺及肽键缩合剂处理.之后两组同时浸于人工唾液中,浸泡2、4、6、8周后扫描电镜观察两组牙本质小管的微观形貌,并使用配对t检验比较8周后两组牙本质小管直径及密度的差异.结果 8周后实验组牙本质小管直径[(2.23 ±0.56)μm]显著低于对照组[(4.20 ±0.67) μm] (P <0.01),牙本质小管密度[(48.9±10.6)个/mm2]也显著低于对照组[(138.0±28.4)个/mm2] (P <0.01).结论 4.0代聚酰胺-胺对敏感牙本质小管有一定的生物矿化及封闭作用,其在治疗牙本质敏感方面存在潜在价值.

Objective To observe and study the biological mineralization of the 4.0th generation of polyamidoamine dendrimer on the sensitive dentinal tubules at different time points in vitro.Methods Thirty-six extracted third molar were cut into 2 mm thick dentin slices to establish sensitive dentin model in vitro.Then the 36 dentine pills were cut into two parts,each using the method of random number table half randomly selected as control group,the other half as experimental group.Samples in the experimental group were treated with the 4.0th generation of polyamidoamine dendrimer synthesized previously and peptide bond condensing agent,while no treatment were applied on samples in the control group.After having immersed in the artificial saliva for 2,4,6,8 weeks respectively,the two groups of dentin slices were observed using scanning electron microscope(SEM) to study their mineralization conditions.Results SEM results showed that the average diameter[(2.23 ±0.56) μm] and density[(48.9 ± 10.6) per square mm] of the dentinal tubules in the experimental group were significantly lower than those in the control group [(4.20 ±0.67) μm,(138.0 ±28.4) per square mm] (P <0.01).Conclusions The result of the experiment implied that the 4.0th generation of polyamidoamine dendrimer has certain effect on the occluding and biomineralization of the sensitive dentinal tubules.Besides,this result also suggested the potential value of the 4.0th generation polyamidoamine dendrimer for the treatment of dentine hypersensitivity.
