目的 探讨血管内皮生长因子(VEGF)与转化生长因子β(TGF-β)基因在用“补肾壮骨合剂”治疗骨缺损过程中的表达及影响.方法 普通级健康家兔48只,其中36只完全随机分成实验组和对照组,各18只,均在无菌条件下取右侧桡骨中下1/3用骨磨钻制成15 mm的骨缺损模型;另外12只家兔处死后,取髂骨制成颗粒状骨,植入以上骨缺损处.实验组用“补肾壮骨合剂”复合饲料喂养.对照组用普通饲料喂养.分别于术后3d和1、2、4、6、8周处死实验组与对照组家兔各3只,取骨缺损处标本,应用聚合酶链反应技术检测VEGF和TGF-β基因的表达.结果 VEGF和TGF-β的目的基因,在校正结果后,以对照组术后1周时测得的基因拷贝值为基础,其他各组与其比值:对照组VEGF的值分别为2.08、1.00、0.88、3.40、1.71、1.17,实验组VEGF的值分别为6.50、2.25、1.86、3.13、3.33、3.31;对照组TGF-β的值分别为0.57、1.00、1.40、1.44、1.59、1.27,实验组TGF-β的值分别为0.58、2.05、2.35、2.59、3.18、2.67.实验组和对照组VEGF基因校正值分别为3.3±1.4比1.7±0.9,差异有统计学意义(t=-18.95,P<0.05),实验组和对照组TGF-β基因校正值分别为2.2±0.9比1.2±0.4,差异有统计学意义(t=-2.594,P<0.05).结论 补肾壮骨合剂能促进VEGF与TGF-β基因的表达.

Objective To investigate the expression and influence of vascular endothelial growth factor (VEGF) and transforming growth factor beta(TGF-β) genes during the treatment of bone defect with setting broken bone mixture (nature copper xuduan gusuibu ground beetle).Methods Thirty-six of forty-eight health adult family rabbits were randomly divided into the experimental group and control group.There were 18 adult family rabbits in every group.Right lower 1/3 radius of the experimental group and the control group were made into a 15 mm standard bone defect model with bone grinding machine.Other twelve rabbits were killed to offer homologous ilium spongy.The experimental group was fed with Chinese drug mixture.Three health adult family rabbits were killed at1st,2nd,4th,6th,8th week after operation in the experimental group.Three health adult family rabbits were killed at1st,2nd,4th,6th,8th week after operation in the control group,Real time Polymerase Chain Reaction (RT-PCR) of vascular endothelial growth factor and transforming growth factor beta genes were investigated.Results After VEGF and TGF-β objective genes were revised for the experimental group and the control group; VEGF valve was 2.08,1.00,0.88,3.40,1.71 and 1.17 in the control group; it was 6.50,2.25,1.86,3.13,3.33,3.31 in the experimental group.TGF-β value was 0.57,1.00,1.40,1.44,1.59 and 1.27 in the control group; it was 0.58,2.05,2.35,2.59,3.18 and 2.67 in the experimental group.Adjusted value of VEGF gene was 3.3 ± 1.4 and 1.7 ± 0.9 in the experimental group and the control group (t =-2.594,P < 0.05).Adjusted value of TGF-β gene was 2.2 ± 0.9 and 1.2 ± 0.4 in the experimental group and the control group (t =-2.594,P < 0.05).Conclusion Setting broken bone mixture can promote the expression level of VEGF and TGF-β gene.
