目的 观察红景天苷对经紫外线A/B (UVA/UVB)辐射的体外培养人皮肤成纤维细胞中Fas和Bcl-2 mRNA表达的影响,探讨红景天苷抑制细胞凋亡与凋亡相关细胞因子的关系.方法 在体外培养人皮肤成纤维细胞并随机分为正常对照组、红景天苷组、UVB组、UVB+红景天苷组、UVA组、UVA+红景天苷组6组,通过反转录聚合酶链反应法检测Fas和Bcl-2的mRNA表达.结果 UVB、UVA辐射体外培养的成纤维细胞后,Fas mRNA表达增加[(0.45±0.04),(0.44±0.04)比(0.13±0.03)],Bcl-2 mRNA表达减弱[0.17±0.01,0.17±0.03比(0.13±0.03)];加入红景天苷后Fas和Bcl-2的mRNA表达分别降低[(0.17±0.04)比(0.45 ±0.04),0.18 ±0.03比0.44±0.04;(0.31±0.02)比(0.17±0.01),(0.28±0.04)比(0.17±0.03)].结论 红景天苷对紫外线引起的Fas和Bcl-2的mRNA表达具有抑制作用,延缓UVB、UVA辐射造成的细胞凋亡过程.

목적 관찰홍경천감대경자외선A/B (UVA/UVB)복사적체외배양인피부성섬유세포중Fas화Bcl-2 mRNA표체적영향,탐토홍경천감억제세포조망여조망상관세포인자적관계.방법 재체외배양인피부성섬유세포병수궤분위정상대조조、홍경천감조、UVB조、UVB+홍경천감조、UVA조、UVA+홍경천감조6조,통과반전록취합매련반응법검측Fas화Bcl-2적mRNA표체.결과 UVB、UVA복사체외배양적성섬유세포후,Fas mRNA표체증가[(0.45±0.04),(0.44±0.04)비(0.13±0.03)],Bcl-2 mRNA표체감약[0.17±0.01,0.17±0.03비(0.13±0.03)];가입홍경천감후Fas화Bcl-2적mRNA표체분별강저[(0.17±0.04)비(0.45 ±0.04),0.18 ±0.03비0.44±0.04;(0.31±0.02)비(0.17±0.01),(0.28±0.04)비(0.17±0.03)].결론 홍경천감대자외선인기적Fas화Bcl-2적mRNA표체구유억제작용,연완UVB、UVA복사조성적세포조망과정.
Objective To explore the effect of salidroside on Fas and Bcl-2 mRNA level in fibroblast apoptosis that ultraviolet radiation B(UVB) and ultraviolet radiation A (UVA) irradiation can induce.Methods The fibroblast was divided into six groups randomly:control group,salidroside group,UVB group,UVA group,UVB group and UVA group treated with salidroside.The mRNA expression levels of Fas and Bcl-2 were determined.Results The expression of Fas and Bcl-2 mRNA Level were obviously increased and decreased after UVB and UVA irradiation compared with the control group [0.45 ± 0.04,0.44 ± 0.04 vs 0.13 ± 0.03 ; 0.17 ± 0.01,0.17 ± 0.03 vs 0.31 ± 0.03].After being treated with salidroside,the expression was decreased and increased [0.17 ± 0.04 vs 0.45±0.04,0.18 ±0.03 vs 0.44 ±0.04; 0.31 ±0.02 vs 0.17 ±0.18,0.28 ±0.04 vs 0.17 ±0.03].Conclusions The UVB/UVA can induce fibroblast apoptosis by decreasing the mRNA expression of Fas(0.45 ±0.04/0.44 ±0.04) and Bcl-2 (0.17 ±0.01/0.17 ±0.03).The salidroside can decrease the level of Fas and Bcl-2 mRNA and fibroblast apoptosis that UVB and UVA irradiation induce.
