目的 观察不同剂量的人脐带胶样组织间充质干细胞(hUMSCs)经静脉移植入大鼠体内对血常规及肝肾功能等指标的影响.方法 ①体外培养人脐带间充质干细胞;利用流式细胞仪鉴定第4代hUMSCs表面抗原的表达;标准试剂盒鉴定第4代hUMSCs向脂肪细胞和成骨细胞分化的潜能;②182~203 g的50只SD大鼠随机分为5组:空白对照组;盐水组;3.5×107/kg干细胞移植为干细胞Ⅰ组;3.5×108/kg干细胞移植为干细胞Ⅱ组;3.5×109/kg干细胞移植为干细胞Ⅲ组;注射后第11天,腹主动脉取血,监测和比较血常规和生化指标,分析比较各项指标的变化和差异.结果 ①第4代hUMSCs特异性表面抗原CD73、CD90和CD105表达强阳性,能分成脂细胞和成骨细胞,比例分别为98.4%和95.7%;②注射干细胞后实验动物全部存活,第11天时实验组24项血常规指标与空白对照组相比差异无统计学意义;③于细胞组血糖较空白对照组明显降低[(8.4±1.6),(7.7±1.6),(7.1±1.6) mmol/L比(9.6±4.9)mmol/L];干细胞组糖化血清白蛋白较空白对照组明显降低[(0.69±0.10),(0.68±0.12),(0.65±0.08) mmol/L比(0.85±0.14) mmol/L];干细胞组的心肌酶水平较空白对照组明显降低[(184±8),(155±24),(144±18)U/L比(223±21) U/L,P<0.05].结论 不同剂量hUMSCs经尾静脉注射入大鼠体内引起血糖、糖化血红蛋白和心肌酶等的降低,且与剂量相关,选择适当剂量的干细胞移植将对临床治疗效果起到重要的作用.

목적 관찰불동제량적인제대효양조직간충질간세포(hUMSCs)경정맥이식입대서체내대혈상규급간신공능등지표적영향.방법 ①체외배양인제대간충질간세포;이용류식세포의감정제4대hUMSCs표면항원적표체;표준시제합감정제4대hUMSCs향지방세포화성골세포분화적잠능;②182~203 g적50지SD대서수궤분위5조:공백대조조;염수조;3.5×107/kg간세포이식위간세포Ⅰ조;3.5×108/kg간세포이식위간세포Ⅱ조;3.5×109/kg간세포이식위간세포Ⅲ조;주사후제11천,복주동맥취혈,감측화비교혈상규화생화지표,분석비교각항지표적변화화차이.결과 ①제4대hUMSCs특이성표면항원CD73、CD90화CD105표체강양성,능분성지세포화성골세포,비례분별위98.4%화95.7%;②주사간세포후실험동물전부존활,제11천시실험조24항혈상규지표여공백대조조상비차이무통계학의의;③우세포조혈당교공백대조조명현강저[(8.4±1.6),(7.7±1.6),(7.1±1.6) mmol/L비(9.6±4.9)mmol/L];간세포조당화혈청백단백교공백대조조명현강저[(0.69±0.10),(0.68±0.12),(0.65±0.08) mmol/L비(0.85±0.14) mmol/L];간세포조적심기매수평교공백대조조명현강저[(184±8),(155±24),(144±18)U/L비(223±21) U/L,P<0.05].결론 불동제량hUMSCs경미정맥주사입대서체내인기혈당、당화혈홍단백화심기매등적강저,차여제량상관,선택괄당제량적간세포이식장대림상치료효과기도중요적작용.
Objective To investigate the effects of exotransplantation of different dose of human umbilical cord mesenchymal stem cell (hUMSCs) on the blood routine and biochemical Indexes in SD rats.Methods Fifty SD rats weighting 182-203 g were divided into 5 groups:Sham,Saline group and different doses of hUMSCs groups (Ⅰ,Ⅱ and Ⅲ).The dosage of hUMSCs was 3.5 × 107/kg,3.5 × 108/kg and 3.5 × 109/kg respectively.hUMSCs in 0.5 ml saline solution were infused through the tail vein.After 11 days of the infusion,the blood samples were drew through the abdominal aorta.The blood routine examination and the biochemical indexes were done in the clinical laboratory.The brain,heart,lung,liver and kidney were collected for HE staining.Results ①The 4th passage of the hUMSCs expressed positively the specific surface antigen such as CD73,CD90 and CD105.The differentiation rate of the hUMSCs towards the osteoblasts and adipocytes were 98.4% and 95.7% ;②Experimental animals all survived after hUMSCs treatment,all blood routine parameters and most biochemical indexes in hUMSCs groups did not show significant difference compared to the control(P < 0.05).③Compared to the control group,the blood glucose,serum glycated albumin,CK valuewere significantly decreased in the hUMSCs groups[(8.4 ± 1.6),(7.7 ±1.6),(7.1 ±1.6)mmol/L vs(9.6±4.9)mmol/L;(0.69±0.10),(0.68 ±0.12),(0.65 ±0.08)mmol/L vs(0.85 ±0.14)mmol/L;(184 ±8),(155 ±24),(144 ± 18)U/L vs(223 ±21)U/L,all P <0.05].Conclusions The different dose of hUMSCs intravenous infusion does not have significant effects on the blood routine results,but the blood glucose and cardiac kinase expression level are decreased significantly in the hUMSCs groups and is related with dosage.The injection of the suitable dosage of hUMSCs is a very important factor for the output of the cellular therapy.
