目的分析2009-2012年引起吉林省手足口病(hand foot and mouth disease,HFMD)流行的肠道病毒71型(EV-A71)的VP1区氨基酸位点的特征性变异位点.方法对2009-2012年分离于吉林省HFMD病例的70株EV-A71分离株的VP1编码区进行逆转录-聚合酶链反应,然后对扩增产物进行核苷酸序列测定,使用Bioedit软件和MEGA 4.0软件分析VP1区氨基酸位点的变异特征以及基因亲缘关系.结果 2009-2012年在吉林省分离到的70株EV-A71均属于C4a基因亚型,其中69株与2008年安徽阜阳的分离株亲缘关系最近,并且这69株病毒在VP1编码区第22位氨基酸位点均由谷氨酰胺转变为组氨酸(Q-H),与2008年安徽阜阳株一致.结论引起2009-2012年吉林省HFMD流行的EV-A71可能来源于安徽阜阳流行株延续传播.

Objective To analyze the variation of amino acid site of VP1 coding region of Enterovirus 71 in Jilin Province in 2009-2012.Methods To choose 70 EV-A71 virus strains isolated from the HFMD cases to do the RT-PCR and sequences of VP1 genes of EV-A71 strains,and analyzed the variation of amino acid site and gene phylogenetic by the software of bioedit and Mega4.0.Results All 70 EV-A71 strains belong to C4a subgenotype in Jilin Province in 2009-2012,among them,69 strains have the closest homology with Fuyang strain of the year of 2008,and the 69 strains have the same variation site from Q to H with that Fuyang strain in the 22th amino acid site in VP1 coding region.Conclusion The EV-A71 which caused HFMD epidemic in Jilin Province in 2009-2012 maybe from continuing transmission of Fuyang strains.
