目的 建立腺病毒3型(HAdV-3)感染HEp-2细胞模型,评价六神丸混悬液对腺病毒感染HEp-2细胞病变的抑制作用,并进一步探讨六神丸在体外抗腺病毒的量效关系.方法 用乙醇回流法处理制备六神丸液.以MTT法计算药物的半数中毒剂量(TC50).通过三种不同给药方式即预防给药、治疗给药及直接灭活给药方式进行体外实验,观察六神丸液对HAdV-3感染HEp-2细胞病变所起的作用,并对药物的量效关系进行分析.结果 六神丸液TC50值为40.48 μg/ml.预防、治疗及直接灭活给药方式均可减轻HAdV-3感染HEp-2细胞的CPE程度,其抗HAdV-3的半数有效浓度(IC50)分别为28.91μg/m1、19.28 μg/m1、8.61 μg/ml,治疗指数(TI)分别为1.4、2.1和4.7,六神丸液对HAdV-3感染HEp-2细胞CPE的抑制作用存在着一定的量效关系.结论 在预防、治疗及直接灭活给药方式下,六神丸均有一定的抗HAdv-3的作用,但直接灭活效果更明显.

목적 건립선병독3형(HAdV-3)감염HEp-2세포모형,평개륙신환혼현액대선병독감염HEp-2세포병변적억제작용,병진일보탐토륙신환재체외항선병독적량효관계.방법 용을순회류법처리제비륙신환액.이MTT법계산약물적반수중독제량(TC50).통과삼충불동급약방식즉예방급약、치료급약급직접멸활급약방식진행체외실험,관찰륙신환액대HAdV-3감염HEp-2세포병변소기적작용,병대약물적량효관계진행분석.결과 륙신환액TC50치위40.48 μg/ml.예방、치료급직접멸활급약방식균가감경HAdV-3감염HEp-2세포적CPE정도,기항HAdV-3적반수유효농도(IC50)분별위28.91μg/m1、19.28 μg/m1、8.61 μg/ml,치료지수(TI)분별위1.4、2.1화4.7,륙신환액대HAdV-3감염HEp-2세포CPE적억제작용존재착일정적량효관계.결론 재예방、치료급직접멸활급약방식하,륙신환균유일정적항HAdv-3적작용,단직접멸활효과경명현.
Objective A model was established by this study to explore anti-adenovirus effect of different administration method of Chinese medicine Liushen pill on adenovirus type 3 (ADV-3) replication in HEp-2 cells.Metods Using ethanol refluxing to prepare Liushen pill suspension.TC50 was calculated by MTT assay.Cell culture was applied to observe the effect of adenovirus type 3(ADV-3) replication through prevention,treatment or direct inactivation of three different drug administration methods.Results Liushen pill suspension was found to be a potential inhibitor of Adv-3 in a concentration(TC50) of 40.48μg/ml in HEp-2 cell culture.The median inhibition concentration(IC50) was 19.28 μg/ml when drug was added after virus infection,and it was 8.61 μg/ml when drug was mixed with virus and added,and it was 28.91 μg/ml when drug was added before virus injection.The therapeutic index (TI) was 1.4,2.1,4.7 repectively.Conclusion The results showed Liushen pill suspension could inhibit the replication of ADV-3 and inactive ADV-3 in vitro.
