目的 观察诱骗受体3( DcR3)小干扰RNA (siRNA)对人结肠癌SW480细胞裸鼠皮下移植瘤DcR3基因表达的影响.方法 建立结肠癌SW480细胞裸鼠皮下移植瘤模型,瘤体注射脂质体与DcR3 siRNA混合物,转染DcR3 siRNA.免疫组织化学法及逆转录-聚合酶链反应(RT-PCR)检测DcR3基因表达.结果 治疗组肿瘤组织免疫组织化学法检测DcR3基因表达阴性;RT-PCR相对吸光度(A)值治疗组(0.12±0.02)明显低于阴性对照组(0.28 ±0.08,P<0.05)和空白对照组(0.27 ±0.05,P<0.05).结论 脂质体与DcR3 siRNA混合物可在结肠癌裸鼠皮下移植瘤内特异性抑制DcR3基因表达.

Objective To observe the effect of decoy receptor 3 ( DcR3 ) small interfering RNA (siRNA) on the expression of DcR3 in athymic mice with subcutaneou transplant tumor of human colon carcinoma SW480 cells.Methods Transplant tumor model was established by subcutaneously transplanting human colon carcinoma SW480 cells into athymic mice.DcR3 siRNA was transfected into transplant tumor in athymic mice and the expression of DcR3 was detected by using immunohistochemistry and reverse transcriptase polymerase chain reaction (RT-PCR).Results The expression of DcR3 gene was negative.The expression of DcR3 gene was obviously lower in treatment group (absorbance:0.12 ± 0.02) than in negative group ( absorbance:0.28 ± 0.08,P < 0.05 ) and blank control group ( absorbance:0.27 ± 0.05,P <0.05 ).Conclusion DcR3 siRNA nediated by liposome distinctly inhibits the expression of DcR3 gene in the transplant tumor in athymic mice with human colon carcinoma SW480 cells.
