目的探讨结直肠癌组织中高尔基磷酸化蛋白3(GOLPH3)表达的临床病理学意义及其与细胞增殖的关系.方法 Western blot检测62例结直肠癌组织和正常结直肠黏膜中GOLPH3的表达,应用免疫组织化学链霉菌抗生物素蛋白-过氧化物酶(SP)法检测哺乳动物雷帕霉素靶蛋白(mTOR)、细胞核增殖抗原(Ki-67)的表达,分析GOLPH3表达与临床病理因素、mTOR表达及Ki-67增殖指数的关系.结果结直肠癌组织中GOLPH3的相对表达量(0.76±0.59)明显高于正常结直肠黏膜的表达量(0.61±0.53,P＜0.05),在细胞分化差、有淋巴结转移或Ⅲ期的结直肠癌组织中GOLPH3的表达量(1.40±1.03、0.97±0.75、0.97±0.75)明显高于高中分化、无淋巴结转移或Ⅰ～Ⅱ期的结直肠癌组织(0.62±0.33、0.57±0.31、0.57±0.31,P＜0.05),此外,GOLPH3的表达与年龄、性别、肿瘤位置、大体类型、肿瘤直径、浸润深度和血癌胚抗原(CEA)水平无明显相关(P＞0.05),mTOR阳性表达的结直肠癌组织中GOLPH3的表达量(0.95±0.73)明显高于mTOR阴性表达的结直肠癌组织(0.54±0.28)；结直肠癌组织中GOLPH3的相对表达量(0.76±0.59)与Ki-67增殖指数(0.46±0.17)呈正相关(r=0.325,P＜0.05).结论 GOLPH3在结直肠癌组织中存在过表达,可促进结直肠癌细胞的增殖.
목적 탐토결직장암조직중고이기린산화단백3(GOLPH3)표체적림상병이학의의급기여세포증식적관계.방법 Western blot검측62례결직장암조직화정상결직장점막중GOLPH3적표체,응용면역조직화학련매균항생물소단백-과양화물매(SP)법검측포유동물뢰파매소파단백(mTOR)、세포핵증식항원(Ki-67)적표체,분석GOLPH3표체여림상병리인소、mTOR표체급Ki-67증식지수적관계.결과 결직장암조직중GOLPH3적상대표체량(0.76±0.59)명현고우정상결직장점막적표체량(0.61±0.53,P＜0.05),재세포분화차、유림파결전이혹Ⅲ기적결직장암조직중GOLPH3적표체량(1.40±1.03、0.97±0.75、0.97±0.75)명현고우고중분화、무림파결전이혹Ⅰ～Ⅱ기적결직장암조직(0.62±0.33、0.57±0.31、0.57±0.31,P＜0.05),차외,GOLPH3적표체여년령、성별、종류위치、대체류형、종류직경、침윤심도화혈암배항원(CEA)수평무명현상관(P＞0.05),mTOR양성표체적결직장암조직중GOLPH3적표체량(0.95±0.73)명현고우mTOR음성표체적결직장암조직(0.54±0.28)；결직장암조직중GOLPH3적상대표체량(0.76±0.59)여Ki-67증식지수(0.46±0.17)정정상관(r=0.325,P＜0.05).결론 GOLPH3재결직장암조직중존재과표체,가촉진결직장암세포적증식.
Objective To explore the expression of Golgi phosphorylation protein 3 (GOLPH3) in colorectal carncer tissues and its correlation with clinicopathological features and cell proliferation.Methods The expression of GOLPH3 protein was examined by using Western blotting in colorectal cancer tissues and colorectal mucosae.Immunohistochemical staining of mammalian target of rapamycin(mTOR) and proliferation cell nuclear antigen (Ki-67) was performed by using the standard streptavidin-peroxidase (SP) technique for the paraffin-embedded sections of 62 colorectal cancers.The relationship between the GOLPH3 expression and the clinicopathological parameters,the mTOR expression or proliferation index was analyzed.Results As compared with normal colorectal mucosae (0.61 ± 0.53),the notably higher level of GOLPH3 protein expression was confirmed in colorectal cancer tissues (0.76 ± 0.59).The expression of GOLPH3 was positively associated with histological differentiation,lymph node metastasis and clinical stage in colorectal cancar (1.40 ± 1.03 vs.0.62 ±0.33),(0.97 ±0.75 vs.0.57 ±0.31),and (0.97 ±0.75vs.0.57 ±0.31) (P ＜0.05).However,there was no correlation between GOLPH3 expression and other clinicopathological parameters such as sex,age,tumor site,tumor depth,length and carcinoembryonic antigen (CEA) l.evel (P ＞ 0.05).The protein level of GOLPH3 expression in mTOR-positive colorectal cancer (0.95 ± 0.73) was significantly higher than in mTOR-negative colorectal cancer (0.54 ± 0.28)(P ＜ 0.05),and protein level of GOLPH3 expression (0.76 ± 0.59) is positively correlated with Ki-67 proliferation index (0.46 ± 0.17) in colorectal cancer via Pearson correlation analysis (r =0.325,P ＜0.05).Conclusion Overexpression of GOLPH3 was found in colorectal cancer tissue,and it may promote cell proliferation through activating mTOR expression.
