目的 探讨视网膜母细胞瘤结合蛋白-6(RBBP6)基因对结肠癌发生、发展的意义及其与结肠癌临床病理特征的关系.方法 应用逆转录-聚合酶链反应(RT-PCR)、实时定量聚合酶链反应(Real-time PCR)检测40例结肠癌患者肿瘤组织、配对的癌旁正常组织RBBP6基因的mRNA表达水平,通过免疫组织化学的方法检测72例结肠癌及其癌旁正常组织RBBP6基因的蛋白表达水平.结果 Real-time PCR结果显示肿瘤组织中RBBP6 mRNA的表达平均值为4.88,正常组织中RBBP6 mRNA的表达平均值为3.49,结肠癌组织RBBP6 mRNA表达水平显著高于癌旁正常组织(P＜0.01).免疫组织化学结果显示RBBP6在正常组织无表达或弱表达[6.9％(5/72)],在肿瘤组织中高表达[68.1％ (49/72)].RBBP6的表达量与区域淋巴结转移(P＜0.05)、肿瘤远处转移(P＜0.001)、肿瘤分期(P＜0.01)和分化(P＜0.05)高度相关.结论 RBBP6基因在结肠癌组织中有较高的表达水平,可能参与结肠癌的发生、发展.

Objective To investigate the association between the expression of retinoblastomabinding protein-6 (RBBP6) gene and the carcinogenesis and development of colon cancer.Methods Use reverse transcription-polymerase chain reaction (RT-PCR) and real-time quantitative polymerase chain reaction (Real-time PCR) to test the expression of RBBP6 mRNA in 40 paired tissues of colon cancer,and use the method of immunohistochemistry to test expression of RBBP6 in 72 paired cancer tissues.Results The results of Real-time PCR shows that the average expression value of RBBP6 mRNA in normal tissues is 3.49 and in cancer tissues is 4.88,the expression value of RBBP6 mRNA in colon cancer tissues is actually higher than the normal tissues (P ＜ 0.01).Immunohistochemical detection of RBBP6 among 72 paired specimens showed that it seldom expressed in normal tissues which only occupies 6.9％ (5/72),while it highly expressed among the cancer tissues which account for 68.1％ (49/72).Increased RBBP6 was significantly associated with lymph node metastasis (P ＜ 0.05),distant metastasis (P ＜ 0.001),tumour histologic grade (P ＜ 0.05) and advanced clinical stage index (P ＜ 0.01).Conclusion The expression of RBBP6 in colon cancer is significantly increased,it is probably related to the carcinogenesis and development of colon cancer.
