目的 筛选结直肠癌患者血浆中差异表达的微小RNA(miRNA).方法 通过TaqMan(R)MicroRNA Array检测7例结直肠癌患者和6例健康体检者血浆中差异表达的miRNA,应用实时荧光定量聚合酶链反应(Real-time PCR)在29例结直肠癌患者和20例健康体检者中进行验证,并分析血浆差异表达的miRNA在结直肠癌中的价值.结果 初步筛选得到16个结直肠癌血浆差异表达的miRNA,Real-time PCR验证得出miR-125b、miR-375、miR-150、miR-206与初筛结果一致且差异有统计学意义(P<0.05).血浆miR-206表达水平对区分结直肠癌患者与健康体检者意义更大,miR-206每增加1个单位,发生结直肠肿瘤风险增加为原来的0.29倍[比值比(OR)=0.29,95%可信区间(CI):0.12 ~0.67],在肿瘤早期就会出现表达水平的明显下降(P<0.01).结论 血浆miR-206与结直肠癌的发展密切相关.

Objective To screen out the plasma miRNA which may be differentially expressed in colorectal cancer.Methods Differential expression of plasma miRNA was detected by using TaqMan(R)MicroRNA Array from 7 patients with colorectal cancer and 6 healthy volunteers.The results were validated in plasma samples of 29 patients with colorectal patients and 20 healthy volunteers by real-time polymerase chain reaction (Real-time PCR).Results Sixteen plasma-specific miRNAs were identified from patients with colorectal cancer,miR-125b,miR-375,miR-150 and miR-206 validated by real-time PCR were consistent with TaqMan(R) MicroRNA Array results and had statistical significant difference (P < 0.05).Furthermore,the expression of plasma miR-206 was more meaningful to distinguish the patients with colorectal cancer from healthy persons.When plasma miR-206 was increased by one unit,the occurrence risk of colorectal cancer was 0.29 [odds ratio(OR) =0.29,95% confidence interval(CI):0.12-0.67] and its expression level was decreased in the early stage of tumor (P < 0.01).Conclusion Plasma miR-206 was expected to be as a non-invasive biomarkerfor early prediction of colorectal cancer.
