目的 观察表没食子儿茶素没食子酸酯(EGCG)、β-羟基-β甲基丁酸盐(HMB)联合应用对小鼠癌症恶病质的治疗作用.方法 于雄性BALB/C小鼠接种小鼠结肠腺癌Colon26(C26)细胞,9d后模型建立.用药7d后,记录小鼠右侧腓肠肌重量、去瘤体质量、血清生化指标、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、C-反应性蛋白(CRP)、肿瘤组织核因子-κB(NF-κB)表达、MAFbx及MURF-1基因表达.结果 安慰剂治疗组、EGCG、HMB、EGCG+ HMB组NF-κB积分吸光度(IA)值分别为21 247.39±5482.31、14 512.99±4654.73、7478.40±2612.45、172.78 ± 173.14,均低于安慰剂治疗组(P <0.05),余各指标亦有不同程度改善(P<0.05).结论 EGCG和HMB联合治疗效果优于草药,其可能通过协同抑制NF-κB表达.

Objective To observe the effect of the combination therapy of Epigallocatechin-3-gallate (EGCG) and β-hydroxy-β-methyl butyrate (HMB) on mouse cancer cachexia model.Methods Male BALB/C mice bearing colon 26 adenocarcinoma for 9 days were served as models of cancer cachexia.Seven days after the treatment,body weight and gastrocnemius muscle were documented.Biochemical indicators,serum interleukin-6 (IL-6),and tumor necrosis factor-α (TNF-α),C-reactive protein (CRP) levels were evaluated.The expression of nuclear factor-κB (NF-tκB) in tumor was detected,and the expressions of MAFbx and MURF-1 genes were valuated.Results IAD levels of NF-κB in placebo treatment group,EGCG group,HMB group and EGCG + HMB group were 21 247.39 ± 5 482.31,14 512.99 ± 4654.73,7 478.40 ±2 612.45,172.78 ± 173.14;respectively.TNF-αlevels in EGCG group,HMB group and EGCG + HMB group were significantly lower than those in placebo treatment group (P < O.05).The expressions of MAFbx and MURF-1 genes were significantly down-regulated in EGCG group,HMB group and EGCG + HMB group compared with those placebo treatment group (P < O.05).Conclusion The combined treatment of EGCG with HMB is better than EGCG or HMB,its mechanism is probably to improve skeletal muscle metabolism by synergistically inhibiting NF-κB pathway.
