目的 分选出HepG2中侧群(SP)细胞并探讨其生物学特性.方法 流式细胞仪分选出SP细胞为实验组,同代常规培养HepG2为对照组,利用非肥胖糖尿病/严重联合免疫缺陷(NOD/SCID)鼠接种实验、黏附实验、Transwell、迁移实验、成球培养实验检测两者生物学特性差异.结果 接种实验显示2×104个SP细胞即能成瘤;两组细胞黏附能力[(16.19±3.68)%比(25.01±13.97)%]和侵袭能力[(20±4)比(16±3)]差异无统计学意义(P>0.05);迁移实验SP细胞穿出细胞数(87±10)比对照组(47±8)多,差异有统计学意义(P <0.05);SP细胞成球数(54 ±9)较对照组(18±3)多(P<0.05).结论 HepG2中SP细胞具有肿瘤干细胞特性,可能富集了肝癌干细胞.
目的 分選齣HepG2中側群(SP)細胞併探討其生物學特性.方法 流式細胞儀分選齣SP細胞為實驗組,同代常規培養HepG2為對照組,利用非肥胖糖尿病/嚴重聯閤免疫缺陷(NOD/SCID)鼠接種實驗、黏附實驗、Transwell、遷移實驗、成毬培養實驗檢測兩者生物學特性差異.結果 接種實驗顯示2×104箇SP細胞即能成瘤;兩組細胞黏附能力[(16.19±3.68)%比(25.01±13.97)%]和侵襲能力[(20±4)比(16±3)]差異無統計學意義(P>0.05);遷移實驗SP細胞穿齣細胞數(87±10)比對照組(47±8)多,差異有統計學意義(P <0.05);SP細胞成毬數(54 ±9)較對照組(18±3)多(P<0.05).結論 HepG2中SP細胞具有腫瘤榦細胞特性,可能富集瞭肝癌榦細胞.

Objective To sort side population cells from hepatocellular carcinoma cell line HepG2 and analze the biological characteristics.Methods Fluorescence-activated cell sorting (FACS) was used to sort side population (SP) cells from HepG2 cells.SP cells served as experimental group and the same generation of HepG2 cells as control group.Non-obese diabetic/severe combined immunodeficiency(NOD/SCID)xenograft transplant experiments,cell-matrix adhesion experiment,Matrigel invasion assay,migration assay and suspension sphere formation assay were used to test the differences in biological characteristics between SP cells and original HepG2 cells.Results NOD/SCID xenograft transplant experiments demonstrated that 2 × 104 SP cells were sufficient to form tumor.There was no significantly difference in adhesion ability [(16.19 ± 3.68) % vs.(25.01 ± 13.97) %] and metastatic potential [(20 ± 4) vs.(16 ± 3)] between the two groups.There was significant difference between experimental group and control group in migrating cells [(87 ± 10) vs.(47 ± 8),(P < 0.05)] and spheres [(54 ± 9) vs.(18 ± 3),(P < 0.05)].Conclusion SP cells sorted from HepG2 cells have cancer stem cell-like property and function,indicating that they may richen in hepatocarcinoma stem cells.
