目的 测定缺血再灌注损伤模型肾组织中肾损伤分子-1(KIM-1)表达,探讨其在反映肾脏热缺血损伤方面的应用价值.方法 建立肾缺血再灌注损伤模型,根据热缺血时间长短随机将大鼠分为对照组(热缺血0 min组)、热缺血15 min组、热缺血30 min组、热缺血45 min组和热缺血60 min组.再灌注8h后,苏木素-伊红(HE)染色观察肾脏病理变化,检测血清肌酐(Cr)值,免疫组织化学法检测肾组织KIM-1表达、Western blot法检测胞质中KIM-1的含量.结果 随着热缺血时间的延长,肾小管肿胀、坏死程度加重,血清Cr值增高,由(32.1±5.7) μmol/L升高至(96.8±9.5) μmol/L,肾组织KIM-1表达增强,累积吸光度(IA)值由372±62升高至40 928±238.45 min热缺血时间内,肾组织KIM-1表达与Cr值呈正相关(r=0.91,P<0.05).结论 大鼠肾组织KIM-1随热缺血时间延长而表达增强,可联合病理学检查,从形态学和功能学两方面反映热缺血损伤对肾脏功能的影响程度.
目的 測定缺血再灌註損傷模型腎組織中腎損傷分子-1(KIM-1)錶達,探討其在反映腎髒熱缺血損傷方麵的應用價值.方法 建立腎缺血再灌註損傷模型,根據熱缺血時間長短隨機將大鼠分為對照組(熱缺血0 min組)、熱缺血15 min組、熱缺血30 min組、熱缺血45 min組和熱缺血60 min組.再灌註8h後,囌木素-伊紅(HE)染色觀察腎髒病理變化,檢測血清肌酐(Cr)值,免疫組織化學法檢測腎組織KIM-1錶達、Western blot法檢測胞質中KIM-1的含量.結果 隨著熱缺血時間的延長,腎小管腫脹、壞死程度加重,血清Cr值增高,由(32.1±5.7) μmol/L升高至(96.8±9.5) μmol/L,腎組織KIM-1錶達增彊,纍積吸光度(IA)值由372±62升高至40 928±238.45 min熱缺血時間內,腎組織KIM-1錶達與Cr值呈正相關(r=0.91,P<0.05).結論 大鼠腎組織KIM-1隨熱缺血時間延長而錶達增彊,可聯閤病理學檢查,從形態學和功能學兩方麵反映熱缺血損傷對腎髒功能的影響程度.

Objective To verify weather kidney injury molecular-1 (KIM-1 could reflect the function of rat kidney undergoing warm ischemia injury (WII).Methods An ischemia reperfusion model was established.Based on the length of warm ischemia time,rats were divided into control group (no WII),15-min WII group,30-min WII group,45-min WII group and 60-min WII group.After reperfusion for 8 h,allograft samples were collected and cut into sections for hematoxylin and eosin (HE) staining,immunohistochemistry and Western blotting,respectively.Blood was collected for further tests.Results With the extension of the warm ischemia time,the kidney tubules became swollen and necrotic.Cr went from (32.1 ±5.7) μmol/L to (96.8 ±9.5) μmol/L,but cumulative absorbance value (IA) of KIM-1 went from 372 ± 62 to 40 928 ± 238.The KIM-1 expression was correlated with Cr (r =0.91,P < 0.01).Conclusion As a biomarker,the expression of KIM-1 get stronger and stranger with the the extension of warm ischemia time.In combination with pathological examination,KIM-1 could reflect function of the kidney under WII.
