目的 观察白细胞介素-17 (IL-17)在糖尿病大鼠肾组织的表达,探讨其参与肾小管-间质病变的机制.方法 55只雄性SD大鼠,随机选择35只以50 mg/kg链脲佐菌素(STZ)制备糖尿病大鼠模型,剩余20只为正常对照组.于2、4周末采用酶联免疫吸附试验(ELISA)方法检测血、尿中IL-17.4周末采用免疫组织化学染色及Western blot检测肾组织中IL-17、α-平滑肌肌动蛋白(α-SMA)及核因子-κB p65(NF-κB p65).结果 糖尿病组大鼠2周末血、尿中IL-17水平分别为(11.51 ±5.62)、(79.21±14.53)ng/L,与对照组比较差异有统计学意义[(8.87±3.35)、(6.75±1.13) ng/L,P<0.01],4周末升高更明显;IL-17在正常对照组肾组织无表达,仅在糖尿病大鼠肾小管和肾间质中高表达.糖尿病大鼠肾组织中IL-17表达与尿中IL-17(r =0.715,P<0.05)及肾组织中α-SMA(r =0.708,P<0.05)、NF-κBp65(r=0.638,P<0.01)表达均呈正相关.结论 IL-17参与糖尿病肾病早期肾小管-间质病变的可能机制为促进α-SMA表达增加.

Objective To investigate the association between the expression of interleukin-17 (IL-17) in renal tubuleinterstitium and diabetic nephropathy.Methods 55 rats were randomly divided into three groups:control group,diabetic rat group.Diabetic rat group was injected 50 mg/kg streptozoticin (STZ).In the 2th,4th week,serum and urine IL-17 were measured with enzyme-linked immunosorbent assay (ELISA).The expression of IL-17,α-smooth muscle actin (α-SMA) and nuclear factor-κB p65 (NF-κB p65) in the kidney were assessed by immunohistochemisty staining and Western blotting.Results Serum and urine IL-17 in diabetic rats were [(11.51 ±5.62),(79.21 ± 14.53) ng/L],which were significantly upreguated as compared with those in control group rats (P < 0.01),which were [(8.87 ± 3.35),(6.75 ± 1.13) ng/L].The expression of IL-17 was only observed in renal tubuleinterstitium of diabetic rats,the expression of IL-17 in renal tubuleinterstitium of diabetic rats was positively correlated with urine IL-17 (r =0.715,P < 0.05),and the expression of α-SMA (r =0.708,P < 0.05)、NF-κB p65 (r =0.638,P < 0.01) in renal.Conclusion The effect of IL-17 on pathogenesis and progression of diabetic nephropathy in diabetic rat might be up-regulating the expression of α-SMA.
