目的 检测人肝细胞癌(HCC)患者分泌型的磷脂酶A2-ⅡA(PLA2G2A) mRNA表达,探讨PLA2G2A与HCC发生发展的关系.方法 采用SYBR Green I荧光实时定量聚合酶链反应(FQ-PCR)方法分别检测51例HCC组织和配对的癌旁肝组织,以及9例正常肝组织中PLA2G2AmRNA表达,以β-肌动蛋白(β-actin)为内参,根据相对定量公式(2-△△CT)计算PLA2G2A mRNA相对表达量,并分析HCC组织中PLA2G2A mRNA表达水平与临床病理特征之间的关系.结果 肝癌组织中PLA2G2A mRNA表达量明显低于配对的癌旁肝组织和正常肝组织,仅是癌旁肝组织的0.2446倍(P<0.01),是正常肝组织的0.2339倍(P<0.01);PLA2G2A mRNA的表达水平和HCC组织的分化程度呈正相关(P<0.05),与HCC患者血清的AFP蛋白的表达量呈负相关(P<0.05);有肝硬化背景的患者HCC组织中PLA2G2A mRNA表达量明显高于无肝硬化背景的患者(P<0.05).结论 PLA2G2A mRNA的表达缺失可能与肝细胞癌的发生和进展有关.

Objective To investigate the correlations between hepatocellular carcinoma (HCC) and phospholipase A2 group ⅡA (PLA2G2A) mRNA expression level.Methods SYBR Green I real-time fluorescent quantitative polymerase chain reaction (FQ-PCR) was used to detect PLA2G2A mRNA expression in 51 HCC tissues and their paired noncancerous liver tissues and 9 normal liver tissues.β-actin gene was used as an internal control and the relative expression level of PLA2G2A mRNA was analyzed using the method of 2-△△CT.The correlation between PLA2G2A mRNA levels in HCC tissues and clinicopathological features was analyzed.Results PLA2G2A mRNA levels in HCC tissues was 0.2446 times lower than that in their paired noncancerous liver tissues (P < 0.01) and 0.2339 times lower than that in normal liver tissues (P < 0.01).PLA2G2A mRNA was positively correlated with tumor differentiation (P < 0.05) and negatively with AFP level (P < 0.05) of HCC.PLA2G2A mRNA levels in HCC tissues underlying liver cirrhosis was higher than those without liver cirrhosis (P < 0.05).Conclusion The downregulation of PLA2G2A may get involved in the tumorgenesis and may be help for diagnosis of HCC.
