目的 观察原发性肝癌(PLC)患者手术前后、介入治疗前后血清Dickkopf-1(DKK1)表达水平的变化.方法 酶联免疫吸附试验法(ELISA)检测30例正常人以及60例PLC患者血清DKK1水平,并比较20例Ⅰ期PLC患者手术前后以及18例Ⅱ期PLC患者介入治疗前后血清DKK1水平.结果 血清DKK1水平与PLC患者临床分期明显相关、与PLC患者性别、年龄无明显相关(P>0.05);PLC患者血清DKK1水平Ⅲ期(11.54±0.80) μg/L明显高于Ⅱ期(9.16±0.73) μg/L、Ⅱ期(9.16±0.73) μg/L明显高于Ⅰ期(6.24±1.12) μg/L、Ⅰ期(6.24 ±1.12) μg/L明显高于正常对照组(3.14 ±0.84) μg/L、两者比较差异有统计学意义(P<0.01);手术后PLC患者血清DKK1水平(5.58 ±1.09) μg/L明显低于手术前(6.24±1.12) μg/L、介入治疗后PLC患者血清DKK1水平(8.81 ±0.80) μg/L明显低于介入治疗前(9.16±0.79)μg/L,两者比较差异有统计学意义(P<0.01).结论 DKK1可能在PLC发展过程中发挥一定的作用.

Objective To investigate the changes of serum Dickkopf-1 (DKK1) in patients with primary liver carcinoma (PLC) before and after operation and interventional therapy.Methods Enzyme linked immunosorbent assay (ELISA) was used to detect the serum DKK1 levels in 60 cases of PLC and 30 cases of normal control subjects.The serum DKK1 levels in 20 cases of stage Ⅰ PLC pre-and post-operation,and 18 cases of stage Ⅱ PLC pre-and post-interventional therapy were compared.Results Serum DKK1 was correlated with clinical stage of patients with PLC,but not with sex and age.The serum DKK1 levels in patients with stage Ⅲ of PLC (11.54 ± 0.80) μg/L were higher than in those with stage Ⅱ (9.16±0.73) μg/L,(P<0.01),those in stage Ⅱ (9.16±0.73) μg/L higher than in stage Ⅰ (6.24 ± 1.12) μg/L,(P < 0.01),and those in stage Ⅰ (6.24 ± 1.12) μg/L higher than in normal controls (3.14 ± 0.84) μg/L.(P < 0.05).The serum DKK1 levels were remarkably reduced in patients with PLC after operation or interventional therapy (P < 0.01).Conclusion These results suggest that the level of DKK1 might play a significant role in the progression of PLC.
