目的 观察Ras同源类似物C(RhoC) mRNA和蛋白在皮肤恶性黑色素瘤组织中的表达,探讨与临床病理特征的关系.方法 应用逆转录聚合酶链反应和Western blot检测32例皮肤恶性黑色素瘤组织中RhoC mRNA和蛋白的表达,以皮肤交界痣和正常皮肤组织作为对照.结果 在皮肤恶性黑色素瘤组织中,RhoC mRNA和蛋白的相对含量分别为0.468±0.230及0.769±0.411,较交界痣及正常皮肤明显升高(P<0.05),在临床Ⅰ、Ⅱ、Ⅲ和Ⅳ期,RhoC mRNA和蛋白的相对含量分别为0.196±0.042、0.411±0.049、0.765±0.091和0.422±0.050、0.763±0.093、1.215±0.076,随临床分期及淋巴结和远处转移RhoC mRNA和蛋白的相对含量逐级升高(P<0.05),但与年龄、性别无明显相关(P>0.05).结论 肿瘤转移相关因子RhoC在皮肤恶性黑色素瘤组织中高表达,且与临床分期及淋巴结和远处转移有关,RhoC可能参与皮肤恶性黑色素瘤的发生发展,且与其侵袭转移过程密切相关.

Objective To investigate the expression of Ras homology C (RhoC) mRNA and protein in cutaneous malignant melanoma and its relationship with clinical pathological features of cutaneous malignant melanoma.Methods The expression of RhoC mRNA and protein was detected by reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blotting in 32 cases of cutaneous malignant melanoma,as well as cutaneous junctional nevus and normal skin tissue were selected as controls.Results The expressions of RhoC mRNA and protein in cutaneous malignant melanoma were 0.468 ± 0.230 and 0.769 ± 0.411,which were higher than those in cutaneous junctional nevus and normal skin tissue (P <0.05).Furthermore,The expressions of RhoC mRNA and protein were 0.196 ± 0.042,0.411 ± 0.049,0.765 ±0.091 and 0.422 ±0.050,0.763 ±0.093,1.215 ±0.076 in the stage Ⅰ,Ⅱ,Ⅲand Ⅳof cutaneous malignant melanoma.The level of RhoC was correlated with increasing of clinical stage or the presence of lymphatic and distant metastases,however it was not correlated with age and gender.Conclusion Overexpression of RhoC,which as a molecular switch to control tumor metastasis,was associated with the carcinogenesis and progression of cutaneous malignant melanoma,and promoted invasion and metastasis in cutaneous malignant melanoma.
