目的检测碳酸酐酶Ⅸ(CA-Ⅸ)在肾细胞癌中的表达,探讨其与临床指标及患者预后的关系.方法采用免疫组织化学(IHC)、逆转录-聚合酶链式反应(RT-PCR)及Western blot等方法检测CA-Ⅸ在63例肾癌组织和63例癌旁组织中的表达.探讨CA-Ⅸ与患者年龄、性别、肿瘤大小、肿瘤TNM分期和转移的关系,并作预后分析.结果 CA-Ⅸ蛋白和基因在肾癌组织中异常高表达,在癌旁组织中低表达或不表达,且CA-Ⅸ的阳性表达随着TNM分期的增高有增高趋势(P＜0.05),并和临床预后密切相关.结论 CA-Ⅸ和肾细胞癌的发生发展密切相关,可能参与肾癌生物学行为的调控.

Objective To detect the expression of carbonic anhydrase-Ⅸ (CA-Ⅸ) in renal cell carcinoma (RCC),and explore its correlation with clinical features and prognosis.Methods Immunohis-tochemistry (IHC),reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blotting were used to detect the expression of CA-Ⅸ in 63 cases of RCC and 63 cases of peritumoral tissues.The relationship of the CA-Ⅸ expression with clinicopathological features and prognosis of RCC was analyzed.Results The expression levels of CA-Ⅸ protein and mRNA in RCC tissues were higher than in peritumoral tissues.With the TNM stage increasing,the expression of CA-Ⅸ was up-regulated (P ＜ 0.05),and there was a close correlation between CA-Ⅸ expression with the clinical prognosis.Conclusion CA-Ⅸ was related to RCC development closely,and may be involved in the regulation of the biological behaviors of RCC.
