目的 观察三磷酸腺苷结合转运家族蛋白4(ABCC4)在胆管癌细胞增殖和转移中的作用.方法 收集2010年6月到2012年6月病理检查确诊胆管癌39例,实时定量聚合酶链反应(Real-time PCR)检测人胆管癌与正常胆管黏膜标本中ABCC4的含量;在人胆管癌细胞株中沉默ABCC4基因;噻唑蓝(MTT)法绘制生长曲线;克隆培养后进行统计.结果 人胆管癌细胞中ABCC4异常高表达,Western blot实验验证在胆管癌细胞中沉默ABCC4基因表达成功,ABCC4基因沉默的HuCCT1细胞增殖速率较对照组明显减缓(P<0.05),克隆实验显示下调ABCC4蛋白表达能够明显降低胆管癌细胞的恶性表征.结论 ABCC4在胆管癌细胞中具有癌基因的功能.
目的 觀察三燐痠腺苷結閤轉運傢族蛋白4(ABCC4)在膽管癌細胞增殖和轉移中的作用.方法 收集2010年6月到2012年6月病理檢查確診膽管癌39例,實時定量聚閤酶鏈反應(Real-time PCR)檢測人膽管癌與正常膽管黏膜標本中ABCC4的含量;在人膽管癌細胞株中沉默ABCC4基因;噻唑藍(MTT)法繪製生長麯線;剋隆培養後進行統計.結果 人膽管癌細胞中ABCC4異常高錶達,Western blot實驗驗證在膽管癌細胞中沉默ABCC4基因錶達成功,ABCC4基因沉默的HuCCT1細胞增殖速率較對照組明顯減緩(P<0.05),剋隆實驗顯示下調ABCC4蛋白錶達能夠明顯降低膽管癌細胞的噁性錶徵.結論 ABCC4在膽管癌細胞中具有癌基因的功能.
목적 관찰삼린산선감결합전운가족단백4(ABCC4)재담관암세포증식화전이중적작용.방법 수집2010년6월도2012년6월병리검사학진담관암39례,실시정량취합매련반응(Real-time PCR)검측인담관암여정상담관점막표본중ABCC4적함량;재인담관암세포주중침묵ABCC4기인;새서람(MTT)법회제생장곡선;극륭배양후진행통계.결과 인담관암세포중ABCC4이상고표체,Western blot실험험증재담관암세포중침묵ABCC4기인표체성공,ABCC4기인침묵적HuCCT1세포증식속솔교대조조명현감완(P<0.05),극륭실험현시하조ABCC4단백표체능구명현강저담관암세포적악성표정.결론 ABCC4재담관암세포중구유암기인적공능.
Objective To investigate the effect of adenosine triphosphate (ATP)-binding cassette transporter sub-family class 4 (ABCC4) on proliferation and metastasis of cholangiocarcinoma cells.Methods There were 39 cases of cholangiocarcinomas from June 2010 to June 2012.Real-time quantitative polymerase chain reaction (Real-time PCR) was used to detect the ABCC4 expression in human cholangiocarcinoma and normal bile duct mucosa.Then ABCC4 gene was silenced in one human cholangiocarcinoma cell line.Growth curve was drawn by using methyl thiazol tetrazolium (MTF) assay.Results The human cholangiocarcinoma cells had a abnormally higher expression of ABCC4.The silencing of ABCC4 was confirmed in cholangiocarcinoma cells by Western blotting.ABCC4 gene-silenced HuCCT1 cells proliferated significantly slower than the control group (P < 0.05).Clone experiments showed that the lowered ABCC4 protein could significantly reduce the malignant phenotypes.Conclusion We speculate that ABCC4 in cholangiocarcinoma cells functions as the role of oncogenes.
