目的 观察细胞窖蛋白-1(Cav-1)在人胰腺癌中的表达,探讨Cav-1在人胰腺癌中的表达及其临床意义.方法 Western blot检测人胰腺癌细胞株(MDA48、MDA28等)和人胰腺导管上皮(HPDE)细胞株中Cav-1蛋白的表达.免疫组织化学检测组织芯片中70例人胰腺癌和10例正常胰腺组织中Cav-1的表达,并分析其与胰腺癌临床病理特征的关系.结果 Western blot结果显示Cav-1在多种人胰腺癌细胞株中高表达.免疫组织化学检测结果显示Cav-1在人胰腺癌组织中高表达,与组织学分级、临床分期及肿瘤转移有关(P<0.01).结论 Cav-1在胰腺癌发生、发展中具有重要作用,与胰腺癌组织学分级、临床分期及肿瘤转移密切相关.

Objective This research is designed to study the expression of Caveolin-1 (Cav-1) in human pancreatic cancer and to analyze its clinical significance.Methods Western blotting were used to detect protein levels of Cav-1 in human pancreatic cancer cell lines (MDA48,MDA28,et al.) and normal human pancreatic ductal epithelial (HPDE) cell line.Tissue microarray-based immunohistochemistry was used to investigate the expression of Cav-1 protein in the 70 primary pancreatic tumor and 10 normal pancreatic tissue specimens.Correlations of Cav-1 expression to histological grade,clinical stage and lymph node or distant metastasis were analyzed.Results Cav-1 was markedly upregulated in most pancreatic cancer cell lines as compared with the HPDE cell line.The results of immunohistochemistry showed that the expression of Cav-1 was upregulated in the pancreatic cancer tissue samples.Increased Cav-1 expression directly correlated with poor level of tumor differentiation,late stage and lymph node metastasis or distant metastasis.Conclusion Increased expression of Cav-1 is associated with poor level of tumor differentiation,late stage and lymph node metastasis or distant metastasis.And Cav-1 may have an oncogenic function in tumorigenesis and progression in pancreatic cancer.
