目的检测肝细胞癌组织中真核肽链释放因子(eRF)3a(eRF3a)/gspt1的表达与临床病理特征的关系,探讨eRF3a/gspt1在肝细胞肝癌发生和进展中的作用.方法应用实时定量聚合酶链反应(ReM-timePCR)、Western blot法检测30例肝细胞癌患者手术切除的癌组织和癌旁组织中eRF3a/gspt1的mRNA和蛋白表达.结果 73.3％肝癌组织中eRF3a/gspt1的mRNA表达显著高于癌旁组织,66.7％肝癌组织中eRF3a/gspt1的蛋白表达显著高于癌旁组织;肝癌组织中eRF3a/gspt1的mRNA表达与性别、年龄、肿瘤大小、甲胎蛋白(AFP)、肿瘤包膜和门脉癌栓差异均无相关(P＞0.05),而与乙肝、病理分级有关;肝癌组织中eRF3a/gspt1的蛋白表达与性别、年龄、肿瘤大小、AFP、肿瘤包膜和门脉癌栓差异均无相关(P＞0.05),而与乙肝、病理分级有关;肝癌组织中eRF3a/gspt1的mRNA表达和蛋白表达之间有相关(P＜0.05).结论 eRF3a/gspt1的高表达可能增加肝癌细胞的蛋白合成,促进肝癌的进展.

Objective To explore the role of eukaryotic peptide chain release factor 3a (eRF3a)/ gspt1 in development and progress of hepatocellular carcinoma (HCC) by detecting the relationship between the expression of eRF3a/gspt1 in HCC and clinicopathological features.Methods The expression of mRNA (73.3％) and protein (66.7％) of eRF3a/gspt1 in 30 cases of HCC tissue was detected by realtime polymerase chain reaction (Real-time PCR) and Western blotting.Thirty cases of paracancerous tissues served as controls.Results The expression of eRF3a/gspt1 mRNA and protein in HCC was significantly higher than the adjacent tissues.The expression of eRF3a/gspt1 mRNA and protein in HCC showed no significant correlation with gender,age,tumor size,alpha fetal protein (AFP),tumor capsule and portal vein tumor thrombus,but significant with hepatitis B and pathological grade.There was correlation between mRNA expression and protein expression of eRF3a/gspt1 in HCC.Conclusion The high expression of eRF3a/gsptl in HCC may enhance protein synthesis,promoting the progress of HCC.
