目的 探讨ZO-1及Claudin-1在胆管癌中的表达并分析其与胆管癌侵袭转移的关系.方法 应用免疫组织化学方法检测Claudin-1和ZO-1在胆管癌、肝转移灶和正常胆管组织中的表达;采用实时定量聚合酶链反应(Real-time PCR)方法检测胆管癌细胞株中Claudin-1及ZO-1基因表达水平.结果 Claudin-1与ZO-1在正常胆管组织中的表达量分别是胆管癌组织中的12.94倍和5.19倍(P<0.01),分别是胆管癌肝转移组织中的125.62倍和60.31倍(P<0.01);Claudin-1与ZO-1在胆管癌组织中的表达与胆管癌是否发生局部或者远处转移密切相关(P<0.05).在3种胆管癌细胞株中,Claudin-1和ZO-1在QBC939和RBE中高表达;在TFK-1中低表达.结论 Claudin-1与ZO-1在胆管癌中的表达与胆管癌的侵袭转移能力密切相关.

Objective To study the expression of zonula occludens-1 (ZO-1) and Claudin-1 in cholangiocarcinoma and analyze the relationship between them with invasion and metastasis potency of cholangiocarcinoma.Methods Immunohistochemistry was employed to detect Claudin-1 and ZO-1 expression in the cholangiocarcinoma,liver metastases and normal bile duct tissues.Real-time quantitative polymerase chain reaction (Real-time PCR) was used to detect the expression of Claudin-1 and ZO-1 in three cholangiocarcinoma cell lines (QBC939,RBE,and TFK-1).Results In bile duct,Claudin-1 and ZO-1 protein was localized on the cell membrane and in the cytoplasm.In the cholangiocarcinoma,Claudin-1 and ZO-1 protein expression was significantly reduced,and located in the cytoplasm.In the cholangiocarcinoma liver metastases,Claudin-1 and ZO-1 were not expressed or very weakly expressed.Semi-quantitative analysis found that the expression of ZO-1 in bile duct was stronger than in cholangiocarcinoma (P < 0.01) and cholangiocarcinoma liver metastases (P < 0.01).The expression of Claudin-1 in bile duct was stronger than in cholangiocarcinoma (P < 0.01) and cholangiocarcinoma liver metastases (P < 0.01).In three cholangiocarcinoma cell lines,Claudin-1 and ZO-1 were relatively strongly expressed in RBE and QBC939,and relatively weakly expressed in TFK-1.Conclusion ZO-1 and Claudin-1 expression is relatively downregulated in cholangiocarcinoma tissues and is undetectable in cholangiocarcinoma liver metastasis tissues.
