目的 观察天然基质金属蛋白酶(MMPs)在大鼠脑动脉瘤模型的作用.方法 健康成年雄性大鼠32只,随机分为高血压组和MMPs干预组(各16只),前者手术造成肾性高血压,后者应用天然性MMP-2、MMP-9干预,分别于术后1、3、5周采集标本,观察脑动脉瘤形成,并检测MMPs血浆浓度及其基因表达水平变化.结果 病理学观察见高血压组有5只大鼠的脑底动脉环有明显动脉瘤形成,干预组结构基本正常.缸浆MMP-2、MMP-9浓度变化:高血压组在术后MMP-2、MMP-9水平逐渐升高,术后3周分别为(105.89±8.24)、(130.79±14.18) mg/L,术后5周分别为(124.34±11.33)、(151.85±16.54) mg/L;MMPs干预组在术后3周分别为(78.97±6.25)、(79.54±7.06) mg/L,术后5周分别为(84.30±7.03)、(86.29±9.27) mg/L,两组MMP-2、MMP-9浓度差异均有统计学意义(P<0.05).免疫组织化学及逆转录-聚合酶链反应(RT-PCR)检测见高血压组在术后3周MMP-2和MMP-9表达明显增加,持续至第5周,干预组仅呈弱阳性表达.结论 血压持续升高并激活MMPs可能是导致脑动脉瘤形成的重要因素.

목적 관찰천연기질금속단백매(MMPs)재대서뇌동맥류모형적작용.방법 건강성년웅성대서32지,수궤분위고혈압조화MMPs간예조(각16지),전자수술조성신성고혈압,후자응용천연성MMP-2、MMP-9간예,분별우술후1、3、5주채집표본,관찰뇌동맥류형성,병검측MMPs혈장농도급기기인표체수평변화.결과 병이학관찰견고혈압조유5지대서적뇌저동맥배유명현동맥류형성,간예조결구기본정상.항장MMP-2、MMP-9농도변화:고혈압조재술후MMP-2、MMP-9수평축점승고,술후3주분별위(105.89±8.24)、(130.79±14.18) mg/L,술후5주분별위(124.34±11.33)、(151.85±16.54) mg/L;MMPs간예조재술후3주분별위(78.97±6.25)、(79.54±7.06) mg/L,술후5주분별위(84.30±7.03)、(86.29±9.27) mg/L,량조MMP-2、MMP-9농도차이균유통계학의의(P<0.05).면역조직화학급역전록-취합매련반응(RT-PCR)검측견고혈압조재술후3주MMP-2화MMP-9표체명현증가,지속지제5주,간예조부정약양성표체.결론 혈압지속승고병격활MMPs가능시도치뇌동맥류형성적중요인소.
Objective To explore the effect of natural matrix metalloproteinases (MMPs) on the cerebral aneurvsm formation in rats.Methods Thirty-two healthy male adult rats were randomly divided into hypertension group and MMPs group (16 cases per group),given operation for establishment of renal hypertension,and natural MMP-2 and MMP-9,respectively.Specimens were collected at 1st,3rd,and 5th week after operation,and the cerebral aneurysms were observed.The dynamic changes of MMP-2 and MMP-9 gene expression were examined.Results Aneurysms were obvious by pathological observation in the bifurcations of Wills arterial ring in 5 rats in hypertension group,but the structures were basically normal in MMPs group.Plasma concentrations of MMP-2 and MMP-9 were (105.89 ±8.24) and (130.79 ±14.18) mg/L at 3rd week,and (124.34 ± 11.33) and (151.85 ± 16.54) mg/L at 5th week in hypertension group; those were (78.97 ±6.25) and (79.54 ±7.06) mg/L at 3rd week,and (84.30 ±7.03) and (86.29 ±9.27) mg/L at 5th week in MMPs group.Between two groups there was significant difference in MMP-2 and MMP-9 concentrations at 3rd week and 5th week after operation (P < 0.05).Immunohistochemistry and reverse transcription-polymerase chain reaction (RT-PCR) revealed that the expression levels of MMP-2 and MMP-9 were significantly increased at 3rd week up to 5th week after operation in hypertension group,but there was only weakly positive expression in MMPs group.Conclusion Sustaining hypertension with plentiful activation of MMPs was an important initating factor of cerebral aneurysm formation,and the mechanism was comparatively complex.
