目的 观察异丙酚联合依达拉奉对大鼠脑局灶性缺血-再灌注损伤后星形胶质细胞胶质纤维酸性蛋白(GFAP)表达的影响.方法 健康雄性Wistar大鼠72只随机分为脑缺血再灌注组(I/R)、依达拉奉干预组(ED)和异丙酚联合依达拉奉组(PE),采用线栓法制作大鼠脑缺血再灌注模型;缺血2h后,设再灌注4、12、24、48 h组,测定大鼠神经功能缺损评分后取脑,采用氯化三苯基四氮唑(TTC)染色法检测大鼠脑梗死体积,采用链霉亲和素-生物素-过氧化物酶复合物(SABC)三步法测定大鼠脑梗死边缘带GFAP表达.结果 I/R组再灌注后4h已出现梗死灶,随着时间的延长,梗死体积逐渐增大,24 h梗死体积最大;ED、PE组梗死边缘带GFAP表达量[(45.10±2.93)、(43.30 ±2.71)个]较IR组表达量[(23.30±2.71)个]增加且表达高峰提前(P<0.05).ED、PE组与I/R组各时间点比较,梗死灶体积[(16.50±0.67)%、(16.80±0.76)%比(20.50±0.78)%]明显缩小(P<0.05),神经功能缺损评分[(2.75±0.61)、(1.77±0.54)比(1.69±0.63)分]明显降低(P<0.05).结论 依达拉奉能提高大鼠脑缺血再灌注后GFAP表达水平,减少脑梗死的面积,减轻脑缺血再灌注后神经功能损伤程度,联合应用异丙酚对其缺血再灌注损伤的保护作用无显著影响.
目的 觀察異丙酚聯閤依達拉奉對大鼠腦跼竈性缺血-再灌註損傷後星形膠質細胞膠質纖維痠性蛋白(GFAP)錶達的影響.方法 健康雄性Wistar大鼠72隻隨機分為腦缺血再灌註組(I/R)、依達拉奉榦預組(ED)和異丙酚聯閤依達拉奉組(PE),採用線栓法製作大鼠腦缺血再灌註模型;缺血2h後,設再灌註4、12、24、48 h組,測定大鼠神經功能缺損評分後取腦,採用氯化三苯基四氮唑(TTC)染色法檢測大鼠腦梗死體積,採用鏈黴親和素-生物素-過氧化物酶複閤物(SABC)三步法測定大鼠腦梗死邊緣帶GFAP錶達.結果 I/R組再灌註後4h已齣現梗死竈,隨著時間的延長,梗死體積逐漸增大,24 h梗死體積最大;ED、PE組梗死邊緣帶GFAP錶達量[(45.10±2.93)、(43.30 ±2.71)箇]較IR組錶達量[(23.30±2.71)箇]增加且錶達高峰提前(P<0.05).ED、PE組與I/R組各時間點比較,梗死竈體積[(16.50±0.67)%、(16.80±0.76)%比(20.50±0.78)%]明顯縮小(P<0.05),神經功能缺損評分[(2.75±0.61)、(1.77±0.54)比(1.69±0.63)分]明顯降低(P<0.05).結論 依達拉奉能提高大鼠腦缺血再灌註後GFAP錶達水平,減少腦梗死的麵積,減輕腦缺血再灌註後神經功能損傷程度,聯閤應用異丙酚對其缺血再灌註損傷的保護作用無顯著影響.
목적 관찰이병분연합의체랍봉대대서뇌국조성결혈-재관주손상후성형효질세포효질섬유산성단백(GFAP)표체적영향.방법 건강웅성Wistar대서72지수궤분위뇌결혈재관주조(I/R)、의체랍봉간예조(ED)화이병분연합의체랍봉조(PE),채용선전법제작대서뇌결혈재관주모형;결혈2h후,설재관주4、12、24、48 h조,측정대서신경공능결손평분후취뇌,채용록화삼분기사담서(TTC)염색법검측대서뇌경사체적,채용련매친화소-생물소-과양화물매복합물(SABC)삼보법측정대서뇌경사변연대GFAP표체.결과 I/R조재관주후4h이출현경사조,수착시간적연장,경사체적축점증대,24 h경사체적최대;ED、PE조경사변연대GFAP표체량[(45.10±2.93)、(43.30 ±2.71)개]교IR조표체량[(23.30±2.71)개]증가차표체고봉제전(P<0.05).ED、PE조여I/R조각시간점비교,경사조체적[(16.50±0.67)%、(16.80±0.76)%비(20.50±0.78)%]명현축소(P<0.05),신경공능결손평분[(2.75±0.61)、(1.77±0.54)비(1.69±0.63)분]명현강저(P<0.05).결론 의체랍봉능제고대서뇌결혈재관주후GFAP표체수평,감소뇌경사적면적,감경뇌결혈재관주후신경공능손상정도,연합응용이병분대기결혈재관주손상적보호작용무현저영향.
Objective To investigate the effect of propofol with edaravone on the expression of glial fibrillary acidic protein (GFAP) after focal brain ischemia-reperfusion injury in rats.Methods The middle cerebral artery (MCA) focal cerebral ischemia-reperfusion rat models were established by thread occlusion method.Rats were randomly divided into focal cerebral ischemia group (I/R),Edaravone-treated group (ED) and Edaravone with Propofol-treated group (PE).After ischemia for 2 h,four subgroups were set up according to the reperfusion time (4,12,24,and 48 h).After measurement of the score of neurological deficits,the infarct volume of brain ischemic damage was estimated by the tovanto transit commission (TTC) staining.Strept avidin-biotin complex (SABC) method was used to detect the expression of brain tissue GFAP.Results The infarction volume was detected at 4 h after focal brain ischemia-reperfusion.Infarction volume became larger as time prolonging.GFAP expression of brain tissue was increased in ED and PE groups [(45.10 ±2.93) vs.(43.30 ±2.71) vs.(23.30 ±2.71)].As compared with control group,the infarction volumes were significantly deflated in ED and PE groups [(16.50 ±0.67)% vs.(16.80 ± 0.76) % vs.(20.50 ± 0.78) %] (P < 0.05),and the scores of neurological deficits were also decreased [(2.75 ±0.61) vs.(1.77 ±0.54) vs.(1.69 ±0.63)] (P<0.05).Conclusion Edaravone can increase the expression of GFAP and cerebral infarction volume,and attenuate neurologic function deficits after cerebral ischemia-reperfusion in rats.
