目的 观察在胃癌大鼠模型胃肠道组织中Krüpple样转录因子6(Klf-6)和核组蛋白-2(NUCB2)及相关基因表达的影响.方法 将Wister大鼠96只分为3组,A组32只,采用黄曲霉素B1(AFB1)对大鼠进行诱导处理3个月,制备胃癌大鼠模型.B组32只,应用AFB1对大鼠进行诱导处理6个月,制备胃癌大鼠模型.C组32只,为正常对照组.用免疫组织化学法检测3组大鼠胃肠道组织中NUCB2和Klf-6的表达水平.逆转录-聚合酶链反应(RT-PCR)技术检测两组大鼠胃肠道组织中NUCB2和Klf-6基因mRNA表达水平,比较其差异.结果 A、B组成功制备了胃癌大鼠模型,但不同诱导时间两组大鼠胃癌发病的程度和发病率不同,差异有统计学意义(P<0.05).Klf-6和NUCB2在B组的表达率高于A组和C组,差异有统计学意义,A组Klf-6和NUCB2表达又高于C组,差异有统计学意义(x2 =3.17、4.95、3.79,P<0.05).RT-PCR结果显示,B组胃肠道组织中Klf-6和NUCB2基因的mRNA表达水平高于A组,A组高于C组,差异有统计学意义(x2=2.98、4.13、3.52,P<0.05).结论 肌氨酸乙酯盐酸盐诱导胃癌模型时间越长,胃癌大鼠胃肠道Klf-6和NUCB2的表达也越强.

Objective To discuss the expression of Krüppel-like factor 6 (Klf-6) and nucleo-bindin-2 (NUCB2) genes in gastrointestinal tract of rats with gastric cancer.Methods Ninety-six adult Wister rats were randomly divided into three groups:32 rats each in groups A and B were drenched intraperitoneally with sarcosine ethyl ester hydrochloride to induce gastric cancer for 3 and 6 months respectively,and 32 rats in group C were injected with normal saline.The gastrointestinal tract was examined by electronic scale at 3rd and 6th month after modeling.The Klf-6 and NUCB2 expression in gastrointestinal tract of rats was observed,and the mRNA levels were evaluated by reverse transcription-polymerase chain reaction (RT-PCR) after the rats were decapitated.The statistical analysis was done by using SPSS 14.0 statistical software.Results All the 96 rats were involved in the analysis of results.Positive rate of Klf-6 and NUCB2 in group A was higher than in groups B and C,and that in group B higher than in group C (x2 =3.17,4.95,and 3.79,P <0.05).RT-PCR revealed that the expression levels of Klf-6 and NUCB2 mRNA in group A were higher than in groups B and C,and those in group B higher than in group C (x2 =2.98,4.13 and 3.52,P < 0.05).Conclusion Combined detection of the expression of Klf-6 and NUCB2 in gastric cancer may give some guideline for the clinical diagnosis,illness evaluation,treatment and prognosis of gastric cancer.
