目的 测定新生血管性青光眼(NVG)患者血清及房水中视黄醇结合蛋白4(RBP4)、中性粒细胞激活肽-78(ENA-78)的水平,探讨RBP4、ENA-78在NVG发生、发展中的作用.方法 选取NVG患者40例(40只眼)作为实验组(A组),原发性慢性闭角型青光眼患者(B组)、老年性白内障患者(C组)各40例(40只眼)作为对照组.采集三组患者血清及房水标本,通过双抗体夹心酶联免疫吸附试验(ELISA)法分别检测血清与房水中RBP4、ENA-78的水平.结果 ①A组房水中RBP4、ENA-78的水平(84±6) mg/L、(249±16) ng/L明显高于B组的(71±7) mg/L、(172±15) ng/L和C组的(59 ±6) mg/L、(85 ±12) ng/L,三组间两两比较,差异均有统计学意义(F=87.16,P<0.01;F=75.39,P<0.01).A组血清中RBP4、ENA-78的水平(57 ±6) mg/L、(184±16) ng/L高于B组的(47±5) mg/L、(135±15) ng/L和C组的(34 ±6) mg/L、(64±12) ng/L,三组间两两比较,差异均有统计学意义(F =73.46,P<0.01F =81.23,P<0.01).②A组房水和血清中RBP4与ENA-78的水平呈正相关(r=0.72,0.67,P均<0.01),其余各组标本中无显著的相关性(P>0.05).结论 NVG患者房水及血清中RBP4、ENA-78的水平明显高于对照组,且房水中二者水平呈明显正相关,提示在NVG病理机制过程中,RBP4、ENA-78作为促血管生成因子,相互促进、相互影响,共同导致了NVG的发生和发展.

Objective To investigate the role of retinol binding protein 4 (RBP4) and epithelial neutrophil-activing peptide-78(ENA-78) in the development of neovascular glaucoma(NVG),by detecting the NVG serum and aqueous humor RBP4 and ENA-78 levels.Methods Forty patients with NVG were selected as experimental group(group A),40 patients with primary chronic angle closure glaucoma patients(group B),and 40 cases of senile cataract patients(group C)as a control group.Three specimens of serum and aqueous humor were collected and the serum and aqueous humor RBP4,ENA-78 levels were measured through the double-antibody sandwich enzyme-linked immuno sorbent assay(ELISA).Results ①The levels of RBP4 and ENA-78 in aqueous humor:group A[(84 ±6) mg/L,(249 ±16) ng/L] were significantly higher than that of group B[(71 ±7) mg/L,(172 ± 15) ng/L] and the group C[(59 ±6) mg/L,(85 ± 12) ng/L].Among the three groups,both the differences of pairwise comparison were significant(F =87.16,P <0.01 ; F =75.39,P <0.01).The levels of serum RBP4,ENA-78:group A[(57 ±6) mg/L,(184 ± 16) ng/L] were higher than that of the group B[(47 ±5) mg/L,(135 ± 15) ng/L] and the group C[(34 ±6) mg/L,(64 ± 12) ng/L].Pairwise comparison among the three groups,the differences were statistically significant(F =73.46,P <0.01; F =81.23,P < 0.01).②NVG group of RBP4 and ENA-78 concentration in serum and aqueous showed a significant positive correlation and statistically significant difference(r =0.72,0.67,P <0.01).The remaining specimens in each group showed no significant correlation (P > 0.05).Conclusions The RBP4,ENA78 levels in the aqueous and serum of patients with NVG were significantly higher than those for the other two groups,and the correlation between the two levels in the aqueous humor was significant positive,suggesting that the RBP4,ENA-78 as an angiogenic factor,promotted and influenced in mutual,led to the occurrence and development of the NVG in the process of NVG pathogenesis.
